目的研究酶法提取土茯苓多糖(SGP)的最佳工艺条件。方法以单因素试验为基础,采用响应面法建立酶解温度、酶解时间和料液比3个因素的回归模型,优化提取工艺。结果分析得最佳提取条件为复合酶(纤维素酶∶果胶酶=1∶2),pH为4.5,酶解温度55℃,酶解时间90 min,料液比1∶40。在此条件下,土茯苓多糖得率为58.2%,多糖提取率为11.7%。结论采用酶解方法能显著提高土茯苓多糖得率。
Objective To study the optimum conditions for enzymatic extraction of Smilax glabra (SGP). Methods Based on the single factor experiment, the regression model was established by using the response surface method to optimize the extraction process by three factors: hydrolysis temperature, enzymolysis time and the ratio of solid to liquid. The results showed that the optimum extraction conditions were enzyme (cellulase: pectinase = 1: 2), pH 4.5, enzymolysis temperature 55 ℃, enzymolysis time 90 min and solid - liquid ratio 1:40. Under these conditions, the yield of Smilax Polysaccharide was 58.2% and the extraction rate of polysaccharide was 11.7%. Conclusion Enzymatic hydrolysis can significantly improve the yield of Smilax china.
