目的研究阿纳其(罗马除虫菊Anacyclus pyrethrum)根提取物对慢性抑郁模型和药物诱发抑郁模型的影响。方法采用慢性温和不可预知应激(CUMS)刺激的方式制备外源性大鼠抑郁模型,通过糖水偏好度、旷场实验、强迫游泳实验、悬尾实验考察大鼠行为学变化;利用Elisa法检测大鼠血清皮质酮的量与海马中5-羟色胺(5-HT)和多巴胺(DA)的量。采用药物诱导方式制备内源性小鼠抑郁模型,分别采用利血平拮抗实验、5-羟色氨酸(5-HTP)诱导甩头实验和育亨宾毒性实验建立内源性小鼠抑郁模型,通过观察小鼠眼睑下垂动物数、运动不能动物数、甩头次数和死亡率,研究阿纳其根提取物抗抑郁作用。结果慢性抑郁模型中,阿纳其根提取物可明显提高CUMS抑郁模型大鼠糖水偏好度、旷场实验中水平穿格次数和直立次数,显著降低强迫游泳和悬尾实验中大鼠的不动时间,同时抑制大鼠血清中皮质酮水平,促进大鼠海马中5-HT和DA水平的升高。药物诱发抑郁模型中,阿纳其根提取物能够减少利血平诱导小鼠的眼睑下垂动物数和运动不能动物数,显著提高5-HTP诱导小鼠的甩头次数,但对育亨宾诱导致死小鼠数量没有影响。结论阿纳其根提取物对慢性抑郁大鼠和药物诱导的抑郁小鼠有一定的抗抑郁作用,其作用机制可能与降低皮质酮和促进5-HT神经功能有关。
Objective To investigate the effects of root extract of Anacyclus pyrethrum on chronic depression and drug-induced depression. Methods Exogenous rat model of depression was prepared by CUMS stimulation. The behavioral changes of rats were investigated by sweet water preference, open-field test, forced swimming test and tail-suspension test. Results In the model of chronic depression, extract of Anatain root can significantly improve the degree of sweetness and water preference, the number of horizontal wears and the upright number in CUMS depression model rats, and significantly reduce the rats’ immobility in forced swimming and tail-suspension experiments Time, while inhibiting the level of corticosterone in rat serum and increasing the level of 5-HT and DA in the hippocampus of rats. In the drug-induced depression model, Anatanjian extract can reduce the number of ptosis and number of animals unable to move in reserpine-induced mice and significantly increase the number of shams in 5-HTP-induced mice, The number of lethal mice has no effect. Conclusion Anataingin extract has some anti-depressant effects on chronic depression and drug-induced depression in mice. Its mechanism may be related to the decrease of corticosterone and the promotion of 5-HT neurological function.
