舒芬太尼预处理对大鼠急性胃黏膜病变组织中TNF-αmRNA与IL-1β mRNA表达的影响

目的观察舒芬太尼预处理对大鼠急性胃黏膜病变过程中胃组织TNF-αmRNA与IL-1βmRNA表达的影响,探讨舒芬太尼是否通过抗炎机制发挥胃黏膜保护作用。方法 24只成年Wistar大鼠随机分成正常组、应激组、舒芬预处理组3组,每组8只,在(20±1)℃水温下,应激组、舒芬预处理组用浸水束缚应激法(WIRS)复制急性胃黏膜病变模型。6h后处死大鼠取胃组织标本,10×解剖显微镜下观察各组胃黏膜损伤指(GI)数,采用RT-PCR技术测定各组胃组织中TNF-αmRNA与IL-1βmRNA的表达变化。结果①与正常组比较,应激组大鼠经WRIS6h后胃黏膜损伤严重,GI明显升高(P<0.01);与应激组比较,舒芬组能显著减轻WRIS后胃粘膜损伤,降低GI(P<0.05)。②与正常组比较,应激组TNF-αmRNA与IL-1βmRNA表达显著上调(P<0.05);与应激比较,舒芬组TNF-αmRNA与IL-1βmRNA表达显著下调。③GI的变化与TNF-αmRNA和IL-1βmRNA的变化呈正相关(r1=0.983,r2=0.993,P<0.05)。结论舒芬太尼预处理能明显下调大鼠急性胃黏膜病变过程中胃组织TNF-αmRNA与IL-1βmRNA的表达,舒芬太尼预处理抑制应激后炎性反应可能为其发挥胃黏膜保护作用的重要机制之一。 Objective To observe the effect of sufentanil preconditioning on the expression of TNF-αmRNA and IL-1βmRNA in gastric mucosa during acute gastric mucosal lesions in rats and to explore whether sufentanil exerts gastric mucosal protective effect through anti-inflammatory mechanism. Methods Twenty-four adult Wistar rats were randomly divided into normal group, stress group and sufefin preconditioning group, with 8 rats in each group. Under the water temperature of (20 ± 1) ℃, Binding stress method (WIRS) replicates acute gastric mucosal lesion. Six hours later, the rats were sacrificed and the gastric mucosal lesion index (GI) was observed under 10 × microscope. The expression of TNF-αmRNA and IL-1βmRNA in gastric mucosa of each group were determined by RT-PCR. Results ① Compared with the normal group, the gastric mucosa of WIS rats were severely damaged and the GI significantly increased (P <0.01). Compared with the stress group, Shufen group could significantly reduce the gastric mucosal damage and the GI (P <0.05). Compared with normal group, the expression of TNF-αmRNA and IL-1βmRNA in stress group were significantly increased (P <0.05); Compared with stress group, the expression of TNF-αmRNA and IL-1βmRNA in Shufen group were significantly down-regulated. ③ The change of GI was positively correlated with the changes of TNF-αmRNA and IL-1βmRNA (r1 = 0.983, r2 = 0.993, P <0.05). Conclusion Sufentanil preconditioning can significantly down-regulate the expression of TNF-αmRNA and IL-1βmRNA in gastric mucosal tissue during the course of acute gastric mucosal lesion in rats. Pretreatment with sufentanil inhibits the inflammatory reaction after gastric mucosal lesion, One of the important mechanisms of action.
