目的探讨雌激素及顺铂对卵巢癌细胞A2780中肽酰基精氨酸脱亚胺酶4(PADI4)表达的作用及其对癌细胞增殖的影响。方法在培养的卵巢癌细胞系A2780细胞中加入不同浓度17β-雌激素作用96 h后(10-12、10-10、10-8、10-6、10-4mmol/L),采用实时定量PCR及Western Blotting检测PADI4表达,并用CCK-8试剂盒检测细胞增殖,选取PADI4的表达变化最明显的雌激素浓度,将A2780细胞分为空白对照组、雌激素组、顺铂组、雌激素加顺铂组(联合组),采用上述方法检测各组PADI4的表达和细胞的增殖情况。结果 1雌激素作用96 h后PADI4表达及细胞增殖随雌激素浓度升高而逐渐降低,但当雌激素浓度升至10-4mmol/L时PADI4表达及细胞增殖率开始升高。2与空白对照组比,联合组和顺铂组培养96 h后PADI4表达均明显降低,且联合组细胞增殖率较雌激素组低。结论雌激素可随浓度不同调控卵巢癌细胞A2780中PADI4的表达和细胞增殖;雌激素对卵巢癌细胞增殖的影响可能与PADI4表达有关。
Objective To investigate the effect of estrogen and cisplatin on the expression of peptidyl-arginine deiminase 4 (PADI4) in ovarian cancer cell line A2780 and its effect on the proliferation of cancer cells. Methods The cultured ovarian cancer cell line A2780 cells were treated with different concentrations of 17β-estrogen for 96 h (10-12, 10-10, 10-8, 10-6, 10-4 mmol / L) And PADI4 were detected by Western Blotting. Cell proliferation was detected by CCK-8 kit. The most obvious estrogen concentration of PADI4 was selected. A2780 cells were divided into blank control group, estrogen group, cisplatin group, estrogen plus cis Platinum group (combined group), using the above method to detect the expression of PADI4 and cell proliferation. Results 1 PADI4 expression and cell proliferation decreased gradually with the increase of estrogen concentration after estrogen treatment for 96 h, but the expression of PADI4 and cell proliferation rate began to increase when estrogen concentration increased to 10-4 mmol / L. 2 Compared with the blank control group, the expression of PADI4 in the combination group and the cisplatin group was significantly decreased after 96 hours of culture, and the cell proliferation rate in the combination group was lower than that in the estrogen group. Conclusion Estrogen may regulate the expression of PADI4 and the proliferation of ovarian cancer cells A2780 with different concentrations. The effect of estrogen on the proliferation of ovarian cancer cells may be related to the expression of PADI4.
