用16个基因型水稻成熟胚在MS和N6培养基上筛选建成悬浮细胞系9个,原生质体再生植株3个。使用一步法或两步法较常规的分步法缩短建成悬浮细胞系时间20~30天。使用“三合一”培养基(即N6大量元素、MS微量元素和B5有机物等)和固液双相培养法较琼脂糖包埋法及液体浅层培养法显著提高原生质体培养的植板率。
Sixteen mature embryos of genotypes of rice were screened on MS and N6 medium to construct 9 suspension cell lines and 3 protoplast regenerated plants. Using the one-step or two-step method, the conventional step-by-step method can shorten the time for establishing the suspension cell line from 20 to 30 days. Compared with agarose embedding method and liquid shallow culture method, “three in one” culture medium (ie N6 macronutrient, MS trace element and B5 organics, etc.) and solid-liquid two-phase culture method significantly increased the plating rate of protoplast culture .
