目的探讨河北省汉族人群D17S30位点的基因频率分布,为法医的基因认定提供依据。方法用聚合酶链式反应(PCR)对河北汉族人群201名无关个体D17S30位点进行扩增,扩增产物采用聚丙烯酰胺凝胶电泳及溴化乙锭染色,依据扩增片段长度确定各等位基因,分别使用POMS软件和自编软件,进行统计学分析和遗传多态性分析,并对10个家系资料进行了分析。结果共检出12种等位基因,45种基因型,基因型分布符合Hardy-Weinberg平衡定律(χ2=10.6872,P=0.7104,df=14)。观测杂合度ho为75.12%,DP、EPP和PIC值分别为0.9611、0.7280和0.8493。该位点呈共显性遗传。结论D17S30位点在河北汉族人群中的基因频率分布具有良好的多态性,适用于该人群的个人识别和亲子鉴定。
Objective To investigate the frequency distribution of D17S30 locus in Han nationality in Hebei province, and to provide basis for forensic gene identification. Methods The D17S30 locus of 201 unrelated individuals in Hebei Han population were amplified by polymerase chain reaction (PCR). Polyacrylamide gel electrophoresis and ethidium bromide staining were used to determine the amplification products. The genes were analyzed by POMS software and self-compiled software respectively for statistical analysis and genetic polymorphism. The data of 10 pedigrees were analyzed. Results A total of 12 alleles and 45 genotypes were detected. The distribution of genotypes conformed to Hardy-Weinberg equilibrium law (χ2 = 10.6872, P = 0.7104, df = 14). The observed heterozygosity ho was 75.12%, DP, EPP and PIC values were 0.9611, 0.7280 and 0.8493, respectively. The site was co-dominant. Conclusion The frequency distribution of D17S30 locus in Hebei Han population has good polymorphism and is suitable for personal identification and paternity testing in this population.
