目的探讨丝线法建立豚鼠不完全性小肠梗阻模型对ICC细胞表达c-kit的影响。方法选取43只成年健康豚鼠随机分成正常A组、假手术B组、模型组(C、D、E)。对模型组豚鼠通过丝线法造成不完全性肠梗阻,3 d后取出丝线并关闭腹腔,分别于术后第1d(C组),第3d(D组),第7d(E组)对小肠收缩力、肠黏膜损伤程度及小肠c-kit含量变化进行测定。结果模型组各项指标的测定与正常组比较均有统计学差异(P<0.05),但E组肠黏膜损伤程度与A组比较无明显差异(P>0.05)。E组小肠收缩力较D组高(P<0.05),肠黏膜损伤程度较D组轻(P<0.01),小肠c-kit含量较D组明显增高(P<0.01)。结论丝线法能建立豚鼠不完全性肠梗阻模型,同时可降低c-kit蛋白表达。
Objective To investigate the effects of silk fibroin on the expression of c-kit in ICC cells induced by incomplete intestinal obstruction in guinea pigs. Methods Forty-three healthy guinea pigs were randomly divided into normal A group, sham operation group B and model group (C, D, E). The model group of guinea pigs caused incomplete intestinal obstruction by thread method. After 3 days, the silk was taken out and the abdominal cavity was closed. The small intestine was contracted on the 1st day (C group), the 3rd day (D group) and the 7th day (E group) Force, intestinal mucosal injury and c-kit intestinal content changes were measured. Results The indexes of the model group were significantly different from those of the normal group (P <0.05), but there was no significant difference between the E group and the A group (P> 0.05). The intestinal contractility of group E was higher than that of group D (P <0.05), and the damage of intestinal mucosa was lighter than that of group D (P <0.01). The content of c-kit of small intestine was significantly higher than that of group D (P <0.01). Conclusion The silk method can establish a model of incomplete intestinal obstruction in guinea pigs and reduce the expression of c-kit protein.
