目的探讨小青龙汤对哮喘小鼠肺组织气道重塑干预作用及对转化生长因子-β1(transforming growth factor-β1,TGF-β1)及白细胞介素-13(Interleukin 13,IL-13)表达的影响。方法 40只雌性小鼠随机分成正常对照组、哮喘模型组、小青龙汤组和强的松组。采用卵蛋白(OVA)致敏2周及OVA滴鼻(共18次)激发6周建立哮喘小鼠模型。小青龙汤组和强的松组分别于每次激发前30 min采用小青龙汤和强的松干预,共6周。HE染色观察各组小鼠肺组织病理变化,Image-Pro Plus 6.0彩色图像分析系统测定支气管壁及平滑肌厚度,免疫组化法和实时定量PCR检测肺组织中TGF-β1和IL-13表达。结果哮喘模型组发生支气管管壁增厚、炎症细胞浸润、平滑肌增生等气道重塑的特征性改变。小青龙汤组和强的松组支气管壁及平滑肌厚度低于哮喘模型组(P<0.05)。免疫组化结果显示:正常组肺组织TGF-β1和IL-13阳性较弱,主要分布在支气管壁;与正常组比较,哮喘模型组TGF-β1和IL-13呈强阳性表达于支气管壁。与哮喘模型组相比较,小青龙汤组和强的松组显著抑制TGF-β1和IL-13基因表达(P<0.05)。结论 TGF-β1和IL-13高表达可能在哮喘气道重塑中起重要作用。小青龙汤改善哮喘小鼠气道重塑可能通过抑制肺组织TGF-β1和IL-13表达而实现。
Objective To investigate the effect of Xiao Qing Long Decoction on the airway remodeling in asthmatic mice and the effect on the expression of transforming growth factor-β1 (TGF-β1) and interleukin-13 (IL-13) Impact. Methods 40 female mice were randomly divided into normal control group, asthmatic model group, Xiao Qing Long Decoction group and prednisone group. The asthma mouse model was established by ovalbumin (OVA) sensitization for 2 weeks and OVA intranasal (18 times) for 6 weeks. Xiaolonglong Decoction group and prednisone group were administered with Xiaoqinglong Decoction and prednisone 30 min prior to each challenge for a total of 6 weeks. The pathological changes of lung tissues in each group were observed by HE staining. The thickness of bronchial wall and smooth muscle were measured by Image-Pro Plus 6.0 color image analysis system. The expression of TGF-β1 and IL-13 in lung tissues were detected by immunohistochemistry and real-time quantitative PCR. Results The characteristic changes of airway remodeling such as bronchial wall thickening, inflammatory cell infiltration and smooth muscle hyperplasia were observed in asthmatic model group. The thickness of bronchus wall and smooth muscle in Xiaoqinglongtang group and prednisone group was lower than that in asthma model group (P <0.05). The results of immunohistochemistry showed that the positive expression of TGF-β1 and IL-13 in normal lung tissue was weak and mainly distributed in the bronchial wall. Compared with normal group, TGF-β1 and IL-13 in asthmatic model group were strongly positive in bronchial wall. Compared with asthma model group, Xiao Qing Long Decoction group and prednisone group significantly inhibited TGF-β1 and IL-13 gene expression (P <0.05). Conclusion High expression of TGF-β1 and IL-13 may play an important role in airway remodeling in asthma. Xiao Qing Long Decoction can improve airway remodeling in asthmatic mice by inhibiting the expression of TGF-β1 and IL-13 in lung tissue.
