不同乙醇浓度提取对总黄酮测定的影响

沙棘为胡颓子科酸棘属的落叶小灌木或乔木,枸杞为茄科植物,两者均有很好的抗氧化作用。据分析它们均含有黄酮类化合物,根据黄酮类化合物与金属离子形成稳定螯合物的性质,可用紫外分光光度法进行总黄酮的测定。而总黄酮的提取可用乙醇作为提取剂,其浓度大致用95%和60%左右。为了解不同浓度提取剂对总黄酮测定结果的影响,本实验分别以95%和60%的乙醇作为提取液,把常用滋补中药枸杞和最近研究得较多的沙棘作为样品,对各自总黄酮含量进行分析测定。材料与方法1.材料:(1)样品沙棘(陕西产)、枸杞(宁夏产);(2)仪器岛津UV-160A紫外分光光度计,万分之一天平;(3)试剂芦丁标准品为Sigma公司产品,其它试剂均为国产分析纯。 Seabuckthorn is a deciduous shrub or arbor in the genus Acanthaceae, and is a solanaceous plant, both of which have good antioxidant activity. According to the analysis, they all contain flavonoids. Based on the properties of flavonoids and metal ions to form stable chelates, the total flavonoids can be determined by UV spectrophotometry. The total flavonoids can be extracted using ethanol as the extractant, the concentration of which is approximately 95% and 60%. In order to understand the effect of different concentrations of extractants on the determination of total flavonoids, 95% and 60% ethanol were used as the extracts in this experiment. Commonly used nourishing herbs and recently researched buckthorn were used as samples, and their total flavonoid content was determined. Analytical determination. Materials and Methods 1. Materials: (1) Samples of Seabuckthorn (Shaanxi) and Chinese Wolfberry (Ningxia); (2) Shimadzu UV-160A Ultraviolet Spectrophotometer, one ten-thousandths of scale; (3) Reagent Rutin standard product for Sigma Other reagents are all domestically produced analytical grades.
