目的比较注射用血栓通(冻干)及其有效组分对缺氧/复氧(H/R)损伤神经元的保护作用。方法体外培养小鼠神经瘤母细胞(Neuro-2a)后,将细胞分为对照组、模型组、给药组(人参二醇皂苷组、人参三醇皂苷组、血栓通组)及配伍组。建立H/R损伤模型。分别以4个质量浓度(50,100,200,400,800mg·L~(-1))的人参二醇皂苷组、人参三醇皂苷组、血栓通组作用细胞24 h和人参二醇、三醇皂苷5个配伍组((100 mg·L~(-1))作用24 h。用CCK-8、试剂盒检测各组中的细胞活力及乳酸脱氢酶(LDH)的漏出量。结果与模型组的细胞存活率为(51.13±1.74)%比较,3个质量浓度(200,400,800 mg·L~(-1))人参二醇皂苷的细胞存活率分别为(24.27±4.88)%,(1.13±0.37)%,(0.93±0.30)%;这3组的LDH漏出率分别为(319.33±31.37)%,(599.32±34.11)%,(879.58±47.20)%,与模型组的(260.97±10.86)%比较,差异均有统计学意义(均P<0.01)。与模型组的细胞存活率为(51.16±6.32)%、LDH的漏出率为(361.20±26.43)%比较,3个质量浓度(200,400,800 mg·L~(-1))人参三醇皂苷的细胞存活率分别为(66.26±3.15)%,60.23±1.93)%,(62.22±4.79)%;这3组的LDH漏出率分别为(295.97±23.62)%,(235.97±43.80)%,(173.33±26.53)%,差异均有统计学意义(均P<0.01)。与模型组的细胞存活率为(50.01±2.83)%、LDH的漏出率为(270±74.24)%比较,3个质量浓度(100,200,400 mg·L~(-1))血栓通组的细胞存活率分别为(58.63±6.07)%,(65.25±8.80)%,(66.38±8.62)%,这3组的LDH漏出率分别为(171.45±13.69)%,(190.55±17.90),(187.07±30.09)%,差异均有统计学意义(均P<0.05),说明药物对损伤的细胞有一定的保护作用。与模型组的细胞存活率为(37.29±3.73)%,第2,5配比组的细胞存活率分别为(46.57±1.27)%,(43.29±2.29)%;第4配比组的细胞存活率(27.92±2.89)%,差异均有统计学意义(P<0.05,P<0.01)。与模型组漏出率为(252.67±9.35)%比较,第2,5配比组的LDH漏出率分别为(211.17±7.73)%,(227.17±13.91)%;第4配比组的LDH漏出率为(295.83±12.77)%,差异均有统计学意义(P<0.05,P<0.01)。结论注射用血栓通(冻干)及其有效组分对H/R损伤的神经元有一定保护作用。 Objective To compare the protective effect of Xueshuantong injection (lyophilized) and its effective components on hypoxic / reoxygenated (H / R) injured neurons. Methods After cultured neuroblastoma cells (Neuro-2a) in vitro, the cells were divided into control group, model group, administration group (Panaxadiol saponin group, Panax ginseng saponin group, Xueshuantong group) and compatibility group. Establish H / R injury model. The cells were treated with 4 concentrations of ginseng diol saponin (50,100,200,400,800mg · L -1), ginseng saponin group and Xueshuantong group for 24 hours, and five groups of ginseng diol and triol saponin (100 mg · L -1) for 24 h.The cell viability and lactate dehydrogenase (LDH) leakage in each group were determined by CCK-8 kit.Results The cell viability in model group was (51.2 ± 4.88)%, (1.13 ± 0.37)%, (0.93 ± 0.37)%, respectively, compared with the control group (51.13 ± 1.74)% at the concentrations of 200,400 and 800 mg · L -1, respectively 0.30)%. The leakage rates of LDH in these three groups were (319.33 ± 31.37)%, (599.32 ± 34.11)% and (879.58 ± 47.20)%, respectively, which were statistically different from those in the model group (260.97 ± 10.86)% (51.16 ± 6.32)%, the leakage rate of LDH was (361.20 ± 26.43)%, and the three mass concentrations (200,400,800 mg · L -1 ), Respectively. The cell viability of the ginsengtriol saponins were (66.26 ± 3.15)% and (60.23 ± 1.93)%, (62.22 ± 4.79)%, respectively. The LDH leakage rates of the three groups were (295.97 ± 23.62)% and ± 43.80)%, (173.33 ± 26.53)%, respectively, with significant difference (all P <0.01). Compared with the model group, the cell survival rate was (50.01 ± 2.83)% and the leakage rate of LDH was (270 ± 74.24)%. The cell survival rates of three groups Respectively, which were (58.63 ± 6.07)%, (65.25 ± 8.80)% and (66.38 ± 8.62)% respectively. The leakage rates of LDH in these three groups were (171.45 ± 13.69)%, (190.55 ± 17.90) and (187.07 ± 30.09) %, The differences were statistically significant (all P <0.05), indicating that the drug on the damaged cells have a protective effect. (37.29 ± 3.73)% in model group and (46.57 ± 1.27)% and (43.29 ± 2.29)% in group 2 and 5, respectively. The cell survival rate in group 4 Rate (27.92 ± 2.89)%, the difference was statistically significant (P <0.05, P <0.01). Compared with the model group (252.67 ± 9.35)%, LDH leakage rates in the second and fifth groups were (211.17 ± 7.73)% and (227.17 ± 13.91)%, respectively. LDH leakage rate in the fourth and fifth groups (295.83 ± 12.77)%, the differences were statistically significant (P <0.05, P <0.01). Conclusion Injection of Xuesaitong (freeze-dried) and its effective components of H / R injury in neurons have a protective effect.
