目的 明确 5 脱氧杂氮胞苷能诱导肝癌细胞凋亡。方法 采用 0 5μmol/L 5 脱氧杂氮胞苷处理肝癌细胞株AMMC 772 1 ,应用流式细胞仪 ,原位末端标记及电镜技术观察经药物处理前后该细胞株的凋亡发生率。结果 经药物处理前后 ,流式细胞仪测得的凋亡发生率分别为 5 8±2 5%和 1 1 1± 3 1 % (P <0 0 5)。末端标记法为 8 3± 4 5%和 1 4 5± 5 2 % (P <0 0 5)。经透射电镜证实有典型的凋亡细胞出现。结论 5 脱氧杂氮胞苷能诱导肝癌细胞SMMC772 1发生凋亡 ,可能与激活了凋亡相关基因的表达有关。
Objective To confirm that 5-deaza-azacytidine can induce hepatocellular carcinoma cell apoptosis. Methods The hepatoma cell line AMMC 772 1 was treated with 0 5 μmol / L 5-deoxy-azacitidine. The apoptosis rate of the cell line was observed by flow cytometry, in situ terminal labeling and electron microscopy. Results The apoptotic rates measured by flow cytometry before and after drug treatment were 58 ± 25% and 11 1 ± 3 1%, respectively (P <0 05). End-labeling was 8 3 ± 45% and 1 45 ± 52% (P <0 05). Transmission electron microscopy confirmed the appearance of typical apoptotic cells. Conclusion 5-deoxy-azacytidine can induce the apoptosis of SMMC772 1 cells, which may be related to the activation of apoptosis-related genes.
