目的:探讨结肠直肠癌组织基质金属蛋白酶-2(matrixmetalloproteinase2,MMP2)和基质金属蛋白酶-9(matrixmetalloproteinase9,MMP9)的表达与肿瘤血管形成的关系。方法:应用免疫组化LSAB法进行研究,抗人MMP2、抗人MMP9和抗人CD31单克隆抗体分别用于检测72例结肠直肠癌手术切除标本MMP2、MMP9的表达情况及肿瘤微血管密度(microvesseldensity,MVD)。结果:72例结肠直肠癌组织中MMP2、MMP9的表达阳性率分别为69.4%和73.6%,DukesA、B、C、D各期的MMP2、MMP9的表达阳性率有递增的趋势(P值分别为0.013和0.027)。肿瘤血管形成与Dukes病理分期也存在一定联系,预后较好的DukesA、B期的MVD值明显低于预后较差的DukesC、D各期(P值<0.001)。MMP2、MMP9的表达情况与肿瘤血管形成的程度显著相关,MMP2、MMP9的表达阳性者的MVD值明显高于MMP2、MMP9的表达阴性者(P值分别为0.033和0.043)。结论:MMP2、MMP9在结肠直肠癌的血管形成中起重要作用,MMP2、MMP9可通过促进肿瘤血管形成影响肿瘤的进展。
Objective: To investigate the relationship between the expression of matrix metalloproteinase-2 (MMP2) and matrix metalloproteinase-9 (MMP9) and tumor angiogenesis in colorectal cancer. METHODS: Immunohistochemical LSAB method was used to study anti-human MMP2, anti-human MMP9 and anti-human CD31 monoclonal antibodies respectively for the detection of MMP2, MMP9 expression in 72 colorectal cancer surgical specimens and microvessel density. MVD). Results: The positive rates of MMP2 and MMP9 expression in 72 cases of colorectal cancer were 69.4% and 73.6% respectively. The positive rates of MMP2 and MMP9 in Dukes A, B, C and D stages were increasing (P values were 0.013 and 0.027). Tumor angiogenesis was also associated with pathological staging of Dukes. The MVD values of Dukes A and B in the better prognosis were significantly lower than those of Dukes C and D with poor prognosis (P < 0.001). The expression of MMP2 and MMP9 was significantly associated with the degree of tumor angiogenesis. The MVD values of MMP2 and MMP9 positive expression were significantly higher than those of MMP2 and MMP9 negative expression (P values were 0.033 and 0.043, respectively). Conclusion: MMP2 and MMP9 play an important role in the angiogenesis of colorectal cancer. MMP2 and MMP9 can influence the progression of tumor by promoting tumor angiogenesis.
