目的采用多波长高效液相色谱(HPLC)梯度洗脱法建立同时测定八宝惊风散中钩藤碱、异钩藤碱、升麻素苷和5-O-甲基维斯阿米醇苷含量的方法。方法采用C18色谱柱(4.6 mm×200 mm,5μm);流速为0.7 ml/min;以乙腈-甲醇(1:4)为流动相A,以0.1%三乙胺溶液为流动相B,梯度洗脱;检测波长:254nm,柱温为30℃。结果钩藤碱、异钩藤碱、升麻素苷和5-O-甲基维斯阿米醇苷质量浓度分别在5.10~102.00μg/ml(r=0.9994)、5.95~119.00μg/ml(r=0.9996)、6.10~122.00μg/ml(r=0.999 9)、5.65~113.00μg/ml(r=0.9998)时与其峰面积呈良好的线性关系;钩藤碱、异钩藤碱、升麻素苷和5-O-甲基维斯阿米醇苷的平均加样回收率分别为97.66%、96.72%、98.99%、99.16%,RSD(n=6)分别为1.32%、0.52%、0.78%、1.56%。结论该方法是一种快速、灵敏、准确的分析方法,可作为八宝惊风散的含量控制方法。
OBJECTIVE To establish a method for simultaneous determination of rhynchophylline, isorhamnetine, cimicifugin, and 5-O-methylvishisainin by multi-wavelength high performance liquid chromatography (HPLC) Method of content. Methods The mobile phase A was consisted of C18 column (4.6 mm × 200 mm, 5 μm), flow rate of 0.7 ml / min, acetonitrile-methanol (1: 4) Off; detection wavelength: 254nm, the column temperature is 30 ℃. Results The concentrations of rhynchophylline, isorhynchophylline, ciclinoid and 5-O-methylvishisainin were respectively 5.10 ~ 102.00μg / ml (r = 0.9994) and 5.95 ~ 119.00μg / ml (r = 0.9996), 6.10-122.00μg / ml (r = 0.999 9), 5.65-113.00μg / ml (r = 0.9998) The average recoveries of glycosides and 5-O-methyl-Wistaroside were 97.66%, 96.72%, 98.99% and 99.16%, respectively, with RSDs of 1.32%, 0.52% and 0.78 %, 1.56%. Conclusion This method is a rapid, sensitive and accurate analytical method, which can be used as the content control method of Babao Xingfeng San.
