目的:探讨棕色脂肪组织(brown adipose tissue,BAT)、解偶联蛋白1(uncoupling protein 1,UCP1)、高脂饮食和中药干预在实验动物能量代谢中的关联性和机制。方法:动物随机分为4组。每组16只。①对照组:普通饲料喂养,d70~d98每天按10 g.kg-1体重生理盐水ig。d98检测刺激性产热,观察2 h肛温变化曲线和温度峰值,计算温度曲线下面积;②阳虚组:d42手术切除肩胛间BAT,余同①组;③脾阳虚组:d49~d98高脂饲料(83%普通饲料,15%甘油三酯,2%胆固醇)喂养;隔日19℃环境喂养,余同②组;④中药组:d70~d98每天按4 g.kg-1体重附子理中汤ig。余同③组。每周测体重1次。以效能体重增长率、效能产热温度峰值、产热温度曲线下面积为参考指标。取BAT测UCP1表达量。结果:①效能体重增长率,脾阳虚组与阳虚组相比体重增长减慢(P<0.01);中药组与脾阳虚组相比,体重增长加快(P<0.01)。②效能产热温度峰值,脾阳虚组与阳虚组相比效能产热温度峰值升高(P<0.01);中药组与脾阳虚组相比,效能产热温度峰值升高(P<0.01);中药组与脾阳虚组相比,效能体重增长率与效能产热温度峰值变化程度呈正相关。③产热温度曲线下面积,脾阳虚组与阳虚组相比产热曲线下面积增加(P<0.05);中药组与脾阳虚组相比,产热温度曲线下面积增加(P<0.01)。效能体重增长率与产热温度曲线下面积变化程度呈正相关。中药组上调UCP1表达量,和阳虚组以及脾阳虚组相比,差异均具有高度显著性(P<0.01)。结论:温阳健脾中药促使高脂饮食转化为BAT并提高其产热活性。温阳健脾中药上调UCP1在BAT中的表达,通过UCP1的解偶联作用,使BAT分解产热而改善阳虚畏寒的症状。
OBJECTIVE: To investigate the association and mechanism of brown adipose tissue (BAT), uncoupling protein 1 (UCP1), high-fat diet and traditional Chinese medicine (TCM) in the energy metabolism of experimental animals. Methods: Animals were randomly divided into 4 groups. 16 in each group. ① control group: normal feed, d70 ~ d98 daily by 10 g.kg-1 body weight saline ig. d98 to detect irritable febrile senescence, 2 h rectal temperature curve and temperature peak were observed, and the area under the temperature curve was calculated; ② Yang deficiency group: d42 surgical removal of interscapular BAT, more than ① group; ③ spleen Yang deficiency group: d49 ~ d98 (83% normal diet, 15% triglyceride, 2% cholesterol); every other day 19 ℃ environment feeding, the same ② group; ④ Chinese medicine group: d70 ~ d98 daily by 4 g.kg-1 body weight of aconitine In the soup ig. Yu Tong ③ group. Body weight measured once a week. With the performance of weight gain rate, the peak value of effective heat production temperature, the area under the curve of heat production temperature as a reference index. Take BAT measurement of UCP1 expression. Results: ① The body weight gain rate of Spleen Yang deficiency group was slower than that of Yang deficiency group (P <0.01). The weight gain of TCM group was faster than that of Spleen Yang deficiency group (P <0.01). ② The peak value of effective heat production temperature, the peak value of effective heat production temperature in spleen-Yang deficiency group was higher than that in Yang deficiency group (P <0.01); Compared with spleen-Yang deficiency syndrome group, the peak value of effective heat production temperature was increased in traditional Chinese medicine group (P < 0.01). Compared with the spleen-yang-deficiency group, the weight gain rate of TCM group was positively correlated with the change of peak value of effective heat-producing temperature. (3) The area under the curve of heat production temperature, the area under the heat production curve in spleen yang deficiency group was significantly higher than that in yang deficiency group (P <0.05); Compared with the spleen yang deficiency syndrome group, the area under the heat production temperature curve was increased (P < 0.01). The rate of change of body weight gain was positively correlated with the change of area under the curve of thermogenic temperature. The up-regulation of UCP1 expression in traditional Chinese medicine group was significantly higher than that in Yangxu group and spleen-Yang deficiency group (P <0.01). Conclusion: Wenyang Jianpi Chinese Medicine can promote high-fat diet into BAT and increase its thermogenic activity. Wenyang Jianpi Chinese Medicine up-regulated the expression of UCP1 in BAT, and through the uncoupling effect of UCP1, BAT produced heat and improved the symptoms of yang deficiency and chills.
