目的 :探讨内毒素 (LPS)致伤后培养人脐静脉内皮细胞 (HUVECs)能量代谢的变化及山莨菪碱、β 七叶皂苷钠干预后对HUVECs能量代谢的影响。方法 :建立体外培养的HUVECs模型 ,致伤因素为LPS ;测定指标为ATP ,ADP及能荷 (EC)。结果 :低浓度LPS能激活内皮细胞 ,随刺激时间的延长 ,培养HUVEC的EC降低 ,用山莨菪碱、β 七叶皂苷钠治疗后各时相点能量和明显增加 ,其中 β 七叶皂苷钠治疗效果优于山莨菪碱。 结论 :LPS对体外培养的HUVEC能量代谢有直接损伤作用 ,β 七叶皂苷钠有一定的抗损伤作用 ,在能量储备方面优于山莨菪碱
Objective: To investigate the changes of energy metabolism in cultured human umbilical vein endothelial cells (HUVECs) induced by endotoxin (LPS) injury and the effects of anisodamin and β-aescinate on the energy metabolism of HUVECs. Methods: The model of HUVECs was established in vitro. The injury factor was LPS. The indexes of measurement were ATP, ADP and energy charge (EC). Results: Low concentration of LPS could activate endothelial cells. With the extension of stimulation time, the EC of cultured HUVECs decreased. After treatment with anisodamine and β-aescinate, the energy and time were significantly increased. Among them, β-aescinate treatment The effect is better than anisodamine. CONCLUSION: LPS can directly damage the energy metabolism of HUVEC cultured in vitro, and β-aescine has a certain anti-injury effect, which is superior to anisodamine in energy reserve
