目的 :提高肿瘤细胞的免疫原性。方法 :将异基因型或同基因型的MHCⅡ类基因转染至小鼠肥大细胞瘤P815中 ,用此细胞接种同系小鼠 ,1个月后给不成瘤的小鼠再接种野生型P815细胞 ,观察小鼠的存活情况。结果 :转同基因型MHCⅡ基因的P815细胞接种同系小鼠后 ,4/ 10不成瘤 ,而异基因型MHCⅡ基因转染组 ,10 / 10不成瘤。 1个月后给不成瘤的小鼠再接种野生型的P815细胞 ,同基因型转染组 2 / 4不成瘤 ,异基因型转染组 7/ 10不成瘤。结论 :研究说明MHCⅡ基因转染具有使肿瘤细胞的致瘤性下降、免疫保护能力提高的作用 ;并且异基因型比同基因型MHCⅡ类基因转染的效果更佳。
Objective: To improve the immunogenicity of tumor cells. Methods: Allogeneic or isogenic MHC class II genes were transfected into mouse mastocytoma P815. The inbred mice were inoculated with this cell line, and the non-tumor-bearing mice were inoculated with wild-type P815 cells one month later. Observe the survival of mice. Results: P105 cells transfected with the same genotype of MHC Ⅱ gene inoculated with syngeneic mice, 4/10 not tumor, and allogeneic MHC Ⅱ gene transfected group, 10/10 not tumor. After 1 month, the non-tumor-bearing mice were inoculated with wild-type P815 cells, 2/4 of the same-genotype transfection group did not form tumor, and 7 of 10 allogenic transfection group did not form the tumor. Conclusion: The study shows that MHC Ⅱ gene transfection has the ability to reduce the tumorigenicity of tumor cells and improve the immune protection ability. And allogeneic genotype is more effective than the same genotype MHC Ⅱ gene transfection.
