为深入了解卡介苗诱导机体产生免疫效应的分子机制,对卡介苗(Bacillus calmettee-guerin,BCG)免疫后小鼠血清特异性抗体与脾脏免疫相关细胞因子表达变化及相关性进行了研究。利用BCG免疫C57BL/6小鼠,采集不同时间点的外周血和脾脏样本。应用间接ELISA方法检测血清中特异性抗体效价,实时荧光定量PCR方法检测脾脏中免疫相关细胞因子表达水平。结果表明:在BCG接种的后期,血清中抗体效价呈极显著的上升趋势(P<0.01);脾脏中Th1型细胞因子白细胞介素-2(Interleukin-2,IL-2)、γ干扰素(Interferon-γ,IFN-γ)的表达水平与对照组相比呈显著性上升(P<0.05),但Th2型细胞因子白细胞介素-4(Interleukin-4,IL-4)的表达水平则明显下降(P<0.05)。这说明BCG免疫后,机体会同时产生细胞免疫应答和体液免疫应答,但免疫后期Th1型细胞因子与体液免疫效应呈正相关,某些Th2型细胞因子则与体液免疫效应呈负相关性。
In order to understand the molecular mechanism of BCG-induced immune response, we studied the changes of immune-related cytokines and the correlation between serum-specific antibodies and spleen in mice after immunization with Bacillus calmettee-guerin (BCG). C57BL / 6 mice were immunized with BCG and peripheral blood and spleen samples were collected at different time points. Serum specific antibody titers were detected by indirect ELISA and the expression of immune related cytokines in spleen were detected by real-time fluorescence quantitative PCR. The results showed that the antibody titer in serum increased significantly at the later stage of BCG inoculation (P <0.01), while the levels of Th1 cytokines such as Interleukin-2 (IL-2), IFN-γ (Interferon-γ, IFN-γ) was significantly increased compared with the control group (P <0.05), but the expression level of Th2 cytokine interleukin-4 (IL-4) Decreased significantly (P <0.05). This shows that after BCG immunization, the body will produce both cellular and humoral immune response, but the post-immune Th1-type cytokines and humoral immune response was positively correlated, while some Th2-type cytokines and humoral immune response was negatively correlated.
