目的:筛选从开口箭不同类型材料中提取DNA的最佳方法。方法:采用十二烷基磺酸钠(SDS)法,改良SDS法,十六烷基三甲基溴化铵(CTAB)法和改良CTAB法4种方法分别对开口箭的新鲜茎、硅胶干燥茎、烘干茎、新鲜叶片和硅胶干燥叶片这5类材料进行DNA提取。从DNA的完整性、纯度、得率等方面对DNA进行比较,确定开口箭不同材料DNA提取的最适方法。结果:采用改良CTAB法提取的硅胶干燥茎和叶片的A260 nm/A280 nm均在1.8~1.9,纯度最高,且完整性较高,提取效果较其他方法更佳;其余材料则均有不同程度的污染,新鲜叶片的A260 nm/A280 nm普遍很低。结论:从硅胶干燥叶片和茎的开口箭材料中更容易提取出纯度高、完整度好的DNA。改良CTAB法是适合开口箭DNA提取的有效方法,通过改良CTAB法从硅胶干燥叶片和茎的开口箭中更容易提取纯度高、完整度好的基因组DNA。
Objective: To screen the best method to extract DNA from different types of materials with open arrows. Methods: Four kinds of methods, sodium dodecyl sulfate (SDS) method, modified SDS method, cetyl trimethyl ammonium bromide (CTAB) method and modified CTAB method, Stems, stems, fresh leaves and silica gel dry leaves these five kinds of materials for DNA extraction. DNA from DNA integrity, purity, yield and other aspects of comparison, to determine the optimum method of DNA extraction of different materials arrows. Results: The A260 nm / A280 nm of the dried stems and leaves of the purified CTAB method were all 1.8-1.9, with the highest purity and higher integrity, and the extraction effect was better than other methods. The remaining materials had different degrees of Pollution, fresh leaves A260 nm / A280 nm generally low. Conclusion: It is easier to extract DNA with high purity and integrity from the dried arrowhead material of the dried leaves and stems of silica gel. The improved CTAB method is an effective method for DNA extraction of open arrows. Genomic DNA with high purity and good integrity can be extracted more easily by modified CTAB method from the openings of dried leaves and stems of silica gel.
