目的:通过胚胎期给药,探讨补肾方对胎儿宫内发育迟缓(IUGR)模型胎鼠己糖激酶(HK)、谷氨酸脱氢酶(GDH)活性的影响。方法:模型组用酒精给孕鼠灌胃导致IUGR模型,各补肾组分别在造模同时予补肾方灌胃,其他组予补气方、补血方、气血双补方灌胃,检测各组胎鼠肝组织HK、GDH活性。结果:IUGR胎鼠HK、GDH活性明显降低(P<0.05);补肾阴方、补血方对IUGR胎鼠HK活性降低均有明显恢复作用(P<0.05);补肾方、补血方、气血双补方均能不同程度恢复IUGR胎鼠GDH活性(P<0.01,P<0.01,P<0.05,P<0.05)。结论:改善IUGR胎鼠肝组织HK、GDH活性可能是补肾方治疗IUGR机理之一,可作为中医“肾主生殖发育、肾为先天之本”和“补母益子”论的依据。
OBJECTIVE: To investigate the effects of Bushen recipe on the activities of hexokinase (HK) and glutamate dehydrogenase (GDH) in fetal rats with intrauterine growth retardation (IUGR) through embryo administration. Methods: IUGR model was induced by intragastric administration of alcohol to pregnant rats in the model group. The rats in each kidney-tonifying group were given intragastric administration of Bushen recipe at the same time. The other groups were fed with invigorating qi, replenishing qi and qi and blood. Fetal rat liver tissue HK, GDH activity. Results: The activities of HK and GDH in IUGR fetus were significantly decreased (P <0.05), and Bushen Yinfang and Buxue Recipe significantly reduced the HK activity in IUGR fetus (P <0.05) All the prescriptions could restore GDH activity of IUGR fetus to varying degrees (P <0.01, P <0.01, P <0.05, P <0.05). CONCLUSION: Improving the activity of HK and GDH in IUGR fetal rat liver tissue may be one of the mechanisms of Bushen recipe for treating IUGR. It can be used as the basis of TCM theory of “Kidney Reproductive Development, Kidney Congenital Benin” and “Supplement” .
