目的:评价N-三甲基壳聚糖(TMC60)包衣脂质体的生物安全性,为其作为肿瘤靶向制剂的载体奠定实验基础。方法:分别采用静脉注射及腹腔注射,对TMC60包衣脂质体的小鼠急性毒性进行了考察,获得LD_(50);同时对其注射刺激性、过敏性及溶血性进行了评价。结果:TMC60包衣脂质体小鼠静脉注射及腹腔注射的LD_(50)和95%置信限(95%CI)分别为224.1(95%CI:178.8~302.6)mg·kg~(-1)和342.3(95%CI:269.7~534.2)mg·kg~(-1),同时在实验剂量时,无注射刺激性、过敏性及溶血性。结论:TMC60包衣脂质体的生物安全性较高,值得进一步研究。
OBJECTIVE: To evaluate the biosafety of NMC-coated liposomes and lay the experimental foundation for its use as a carrier for tumor targeting agents. Methods: The acute toxicity of TMC60 coated liposome mice was investigated by intravenous injection and intraperitoneal injection, respectively. LD_ (50) was obtained. The irritation, allergic and hemolytic injections were also evaluated. Results: The LD_ (50) and 95% CIs of TMC60 coated liposome mice were 224.1 (95% CI: 178.8 ~ 302.6) mg · kg ~ (-1) And 342.3 (95% CI: 269.7 ~ 534.2) mg · kg ~ (-1), respectively. At the same time, there was no injection irritation, allergic and hemolytic at the experimental dose. Conclusion: The bio-safety of TMC60 coated liposomes is high and worth further study.
