PPARα是核受体超家族成员之一,除刺激脂肪酸的氧化作用和改善脂蛋白代谢,PPARα也发挥多效的抗炎和抗增殖效应,防止巨噬细胞中胆固醇堆积引起的促动脉粥样硬化作用。人工合成的PPARα激动剂贝特类药物,用于治疗代谢综合征病人的血脂障碍,减轻心血管疾病及其并发症,减少心肌坏死面积,改善缺血后心肌收缩功能障碍。现将关注PPARα在代谢疾病及其相关的血管病理方面的作用及机制。
PPARα is a member of the nuclear receptor superfamily. In addition to stimulating fatty acid oxidation and improving lipoprotein metabolism, PPARα also exerts potent anti-inflammatory and antiproliferative effects against the pro-atherogenic effects of cholesterol accumulation in macrophages effect. Synthetic PPARα agonist fibrates for the treatment of dyslipidemia in patients with metabolic syndrome, reduce cardiovascular disease and its complications, reduce myocardial necrosis area, improve myocardial ischemic postconditioning dysfunction. We will now focus on the role and mechanism of PPARα in metabolic diseases and their associated vascular pathology.
