本文提出了能在大量中药存在下,测定珍菊降压片中盐酸可乐宁及双氢氯噻嗪的含量,方法准确精密。片剂中赋形剂及填料不干扰测定,适合工厂质量控制分析用。盐酸可乐宁的测定法为先加碱使可乐宁游离后,用氯仿提取,经碱洗除去杂质,再用pH4枸橼酸缓冲液提取,然后用酸性染料法与溴酚蓝试剂形成离子对络合物,用氯仿萃取后,在403nm波长处测定溶液吸收度,盐酸可乐宁浓度在0.8~4PPM范围内符合比尔定律,标准偏差98.40±3.7。双氢氯噻嗪的测定是以水——丙酮(1:4)提取主药后,以氢氧化钠加热水解,水解物经亚硝酸重氮化后,与二盐酸基乙二胺偶合产生有色物,可在518nm波长处测定溶液吸收度。双氢氯噻嗪浓度在1~5PPM范围内符合比尔定律。标准偏差95.27±3.19。
This article proposes to determine the content of clonidine hydrochloride and hydrochlorothiazide in Zhenju Jiangya Tablets in the presence of a large number of traditional Chinese medicines. The method is accurate and accurate. Tablet excipients and fillers do not interfere with the assay and are suitable for factory quality control analysis. The determination of clonidine hydrochloride was performed by first adding alkali to make clonidine free, extracting with chloroform, removing impurities by caustic washing, and then extracting with pH 4 citric acid buffer, and then forming an ion-pairing network with bromophenol blue reagent by an acid dye method. After the mixture was extracted with chloroform, the absorbance of the solution was measured at a wavelength of 403 nm. Beerine’s law was observed within a range of 0.8-4 PPM, and the standard deviation was 98.40±3.7. The hydrochlorothiazide was determined by extracting the main drug with water-acetone (1:4) and then hydrolyzing with sodium hydroxide. The hydrolysate was diazotized with nitrous acid and then coupled with ethylenediamine dihydrochloride to give a colored product. The absorbance of the solution was measured at a wavelength of 518 nm. The hydrochlorothiazide concentration was in the range of 1 to 5 PPM in accordance with Beer’s law. The standard deviation is 95.27±3.19.
