肿瘤的生长,转移等过程均与蛋白水解酶有关,在恶性细胞中蛋白水解酶升高,它能降解基质和结缔组织,从而有利于肿瘤的生长和转移.我们研究了机体内存在的(约占血清总蛋白鼠的3%)广谱蛋白水解酶抑制剂甲_ 2巨球蛋白(α_2M)对肿瘤生长、转移的影响.离体实验采用SGC-7901胃腺癌细胞和B16黑色素瘤细胞,整体动物(小鼠)接种用Lewis肺癌细胞,实验内容有:用核孔滤膜法观察癌细胞的迁移能力;计数癌细胞的集落形成;观察癌细胞的生长表现; 以H~3-TdR标记癌细胞观察其侵袭粘附血管内皮细胞的能力;观察接种癌后动物的存活情况;接种部位癌块的生长体积;检查癌肺部转移灶的数量;测定组织蛋白酶D和超氧化
Tumor growth, metastasis and other processes are related to proteolytic enzymes. In malignant cells, proteolytic enzymes are elevated. It can degrade the matrix and connective tissue, which is conducive to the growth and metastasis of tumors. We have studied the existence of the body (about 3% of serum total protein mice) The effect of a broad-spectrum proteolytic enzyme inhibitor alpha macroglobulin (α_2M) on tumor growth and metastasis. In vitro experiments were performed using SGC-7901 gastric adenocarcinoma cells and B16 melanoma cells. The animals (mice) were inoculated with Lewis lung cancer cells. The experimental contents were: nuclear membrane filter method was used to observe the migration ability of cancer cells; the colony formation of cancer cells was counted; the growth performance of cancer cells was observed; and H~3-TdR was used to mark cancers. The cells were observed for their ability to invade adherent vascular endothelial cells; the survival of the animals after inoculation with cancer was observed; the growth volume of tumor blocks at the site of inoculation; the number of cancerous lung metastases was examined; and cathepsin D and superoxidation were measured.
