目的:观察体外诱导骨髓间充质干细胞向肌成纤维细胞分化及一贯煎的影响。方法:骨髓间充质干细胞分离、培养和鉴定,取第3代细胞加入转化生长因子(TGF-β1 5μg.L-1)诱导定向分化。骨髓间充质干细胞随机分为正常组、TGF-β1诱导组,TGF-β1诱导加一贯煎组,2周后通过免疫荧光、实时定量PCR法检测骨髓间充质干细胞分化情况及一贯煎的影响。结果:骨髓间充质干细胞鉴定结果显示CD44强阳性表达,CD29弱阳性表达。TGF-β1诱导后,免疫荧光及实时定量PCR均显示TGF-β1诱导组细胞α-SMA阳性表达,一贯煎组细胞α-SMA表达显著降低,差异有统计学意义(P<0.05)。结论:经体外诱导骨髓间充质干细胞可以向肌成纤维细胞分化,一贯煎可阻止骨髓间充质干细胞向肌成纤维细胞分化。
OBJECTIVE: To observe the effect of inducing bone marrow mesenchymal stem cells to differentiate into myofibroblasts in vitro and consistently. METHODS: BMSCs were isolated, cultured and identified. The third generation cells were induced to differentiate by adding transforming growth factor (TGF-β1 5 μg.L-1). Bone marrow-derived mesenchymal stem cells were randomly divided into normal group, TGF-β1-induced group and TGF-β1-induced group. The expression of BMSCs was detected by immunofluorescence and real-time quantitative polymerase chain reaction . Results: The results of bone marrow mesenchymal stem cells showed strong positive expression of CD44, weak positive expression of CD29. After induction of TGF-β1, the expression of α-SMA in TGF-β1-induced cells was significantly increased by immunofluorescence and real-time quantitative PCR. The α-SMA expression was significantly decreased in the group treated with TGF-β1 (P <0.05). CONCLUSION: Bone marrow-derived mesenchymal stem cells can differentiate into myofibroblasts in vitro and consistently prevent bone marrow mesenchymal stem cells from differentiating into myofibroblasts.
