目的研究并优化荷叶生物碱的提取工艺。方法采用酸提碱沉提取结合有机溶剂萃取纯化的方法从荷叶中提取生物碱。并通过单因素试验和正交试验,研究pH值、提取温度、提取时间、固液比等条件对荷叶生物碱提取的影响,优化工艺提取条件。结果荷叶生物碱的最佳提取工艺:提取温度90℃,pH值2.5,提取时间2 h,固液比1∶15。采用此条件提取的荷叶生物碱纯度为73.4%。提取温度、pH值对提取率影响显著。结论该荷叶生物碱的提取纯化工艺简单,成本低,提取效率高。
Objective To study and optimize the extraction process of lotus leaf alkaloids. Methods Extract alkaloids from lotus leaves by acid extraction and precipitation combined with organic solvent extraction and purification. The effects of pH value, extraction temperature, extraction time, solid-liquid ratio and other conditions on the extraction of alkaloids from lotus leaf were studied by single factor test and orthogonal experiment, and the extraction conditions were optimized. Results The best extraction process of alkaloids from lotus leaf was extraction temperature 90 ℃, pH value 2.5, extraction time 2 h and solid-liquid ratio 1:15. The purity of lotus leaf alkaloids extracted by this condition was 73.4%. Extraction temperature, pH value of the extraction rate significantly. Conclusion The lotus leaf alkaloids extraction and purification process is simple, low cost, high extraction efficiency.
