为探明海蜇养殖群体的种质情况,了解海蜇自然群体与养殖群体的遗传多样性水平,本研究基于Illumina高通量测序获得的海蜇转录组数据,利用MISA软件筛选出5088个微卫星位点,挑选出100个微卫星位点设计引物,58个位点扩增出目的条带,其中25对微卫星引物具有多态性,并对江苏自然群体和辽宁养殖群体进行遗传多样性分析。结果显示,江苏自然群体和辽宁养殖群体的平均等位基因数分别为3.120、2.360,有效等位基因数分别为2.112、1.738,期望杂合度平均值分别为0.495、0.378,多态信息含量均值分别为0.420、0.311,香农信息指数均值分别为0.823、0.593。统计结果显示,两个群体的遗传多样性处于中度多态性水平,自然群体的遗传多样性水平稍高于养殖群体。本研究的结果为海蜇遗传改良、遗传连锁图谱构建和种质资源评价等方面的研究提供理论基础和参考资料。
In order to find out the germplasm resources of jellyfish breeding population and understand the genetic diversity of jellyfish natural population and cultured population, based on the data of jellyfish transcriptome obtained from Illumina high-throughput sequencing, 5088 microsatellite loci , Selected 100 microsatellite loci design primers, 58 loci amplified target bands, of which 25 pairs of microsatellite primers with polymorphisms, and Jiangsu Province, Liaoning Province and Liaoning Province population breeding population genetic diversity analysis. The results showed that the average number of alleles of Jiangsu natural population and Liaoning farming population were 3.120 and 2.360 respectively, the number of effective alleles was 2.112 and 1.738 respectively, and the average expected heterozygosity was 0.495 and 0.378 respectively. The mean of polymorphic information content 0.432, 0.311, Shannon information index mean were 0.823,0.593. The statistical results show that the genetic diversity of the two groups is at a moderately polymorphic level, and the genetic diversity of the natural groups is slightly higher than that of the cultured groups. The results of this study provide the theoretical basis and reference materials for the genetic improvement of jellyfish, genetic linkage map construction and germplasm resources evaluation.
