目的 研究神经生长因子 (NGF)和溴隐亭 (BC)对体外培养的垂体催乳素 (PRL)分泌腺瘤细胞的激素分泌量和增殖的影响。方法 每例体外培养的垂体PRL分泌腺瘤细胞被分成 4组 :对照组、NGF组、BC组和NGF +BC组 ,对不同组进行不同的药物干预 ,观测培养液中激素分泌量和3H 胸腺嘧啶脱氧核苷 (TdR)摄取率的改变。结果 在 8例培养成活的垂体PRL分泌腺瘤细胞中 ,与对照组相比 ,NGF对PRL分泌量的抑制作用不明显 ,但有 3例细胞3H TdR摄取率明显降低(P <0 .0 1) ;BC对 5例细胞PRL分泌量有明显的抑制作用 (P <0 .0 1) ,3例细胞3H TdR摄取率明显下降 (P <0 .0 1) ,3例细胞3H TdR摄取率下降非常明显 (P <0 .0 0 1) ;二者联合应用 ,对PRL分泌量和3H TdR摄取率的抑制作用更加明显 (P <0 .0 0 1)。结论 NGF可增强BC对垂体PRL分泌腺瘤细胞激素分泌的抑制作用 ,并对肿瘤细胞的增殖产生抑制作用
Objective To investigate the effects of nerve growth factor (NGF) and bromocriptine (BC) on hormone secretion and proliferation of cultured pituitary prolactin (PRL)-secreting adenoma cells. Methods Each in vitro cultured pituitary PRL-secreting adenoma cell was divided into 4 groups: control group, NGF group, BC group and NGF +BC group. Different groups were given different drug interventions to observe hormone secretion and 3H thymus in culture medium. Changes in the uptake rate of pyrimidine deoxynucleosides (TdRs). Results In the eight cultured pituitary PRL-secreting adenoma cells, compared with the control group, the inhibitory effect of NGF on the secretion of PRL was not obvious, but the 3H TdR uptake rate was significantly reduced in 3 cases (P < 0.01). BC significantly inhibited the secretion of PRL in 5 cases (P < 0.01), the 3H TdR uptake rate in 3 cases was significantly decreased (P < 0.01), and the rate of 3H TdR uptake was decreased in 3 cases. It was very obvious (P < 0.01); combined use of the two, the inhibition of PRL secretion and 3H TdR uptake rate was more pronounced (P < 0.01). Conclusion NGF can enhance the inhibitory effect of BC on cytokine secretion of pituitary PRL secreting adenomas and inhibit the proliferation of tumor cells.
