红芪多糖1联合顺铂对肺癌A549细胞凋亡的协同作用及其机制探讨

目的:探讨红芪多糖1对顺铂致肺腺癌A549细胞凋亡的增效作用。方法:MTT法检测不同浓度的红芪多糖1、顺铂及二者联合用药对肺腺癌A549细胞生长的抑制作用,FCM法检测细胞凋亡率及细胞周期分布,实时荧光定量PCR法检测Bcl-2和Bax的mRNA表达情况。结果:不同浓度的红芪多糖1对A549细胞的生长均有抑制作用,抑制率与浓度和时间呈正相关。红芪多糖1联合顺铂可使A549细胞凋亡率较单用顺铂组凋亡率明显增加(P<0.05),且细胞周期阻滞在G1和S期。不同浓度的红芪多糖1可降低Bcl-2蛋白表达(P<0.05),增加Bax蛋白表达(P<0.05),具有浓度依赖性;而且红芪多糖1联合顺铂处理对Bcl-2和Bax蛋白表达的抑制或促进作用均较单用顺铂组明显增强(P<0.05),具有协同作用。结论:红芪多糖1可提高顺铂诱导的A549细胞凋亡率,其机制可能与降低Bcl-2/Bax mRNA比例有关。 Objective: To investigate the synergistic effect of Radix Hedysari Polysaccharides 1 on the apoptosis of lung adenocarcinoma A549 cells induced by Cisplatin. Methods: MTT assay was used to detect the inhibitory effects of Radix Hedysari Polysaccharide 1, cisplatin and their combination on the growth of lung adenocarcinoma A549 cells. FCM was used to detect the apoptosis rate and cell cycle distribution. Real-time fluorescence quantitative PCR was used to detect the expression of Bcl- -2 and Bax mRNA expression. Results: Different concentrations of Radix Hedysari 1 inhibited the growth of A549 cells. The inhibition rate was positively correlated with the concentration and time. Compared with Cisplatin alone, the apoptosis rate of A549 cells was significantly increased (P <0.05), and cell cycle arrest was at G1 and S phase. Different concentrations of Radix Hedysari 1 could decrease the expression of Bcl-2 protein (P <0.05) and increase the expression of Bax protein (P <0.05) in a concentration-dependent manner. Inhibition or promotion of protein expression were significantly enhanced (P <0.05) compared with cisplatin alone, with synergistic effect. Conclusion: Hedysari polysaccharide 1 can increase the apoptosis rate of A549 cells induced by cisplatin, which may be related to the decrease of the ratio of Bcl-2 / Bax mRNA.
