目的 研究钙拮抗剂尼卡地平 (Nicardipine ,NC)对恶性胶质瘤细胞系U2 5IMG的细胞生长效应。方法 在无血清培养基中 ,予不同剂量的NC、EGF及NC和EGF (10ng ml)干预 ,应用MTT比色法观察它们的细胞生长效应。结果 EGF对U2 51MG细胞表现出剂量依赖性生长刺激作用 ,10ng ml时接近最大效应 ,与 10 %小牛血清的促生长效应相仿 (P >0 0 5) ;NC抑制U2 51MG细胞生长 ,并完全阻断EGF的效应。结论 尼卡地平能抑制胶质瘤细胞生长并完全阻断EGF的生长刺激作用 ,为临床上治疗胶质瘤提供一条新的线索
Objective To investigate the effect of Nicardipine (NC), a calcium antagonist, on the growth of human glioblastoma cell line U2 5IMG. Methods Different doses of NC, EGF, NC and EGF (10ng ml) were used in serum-free medium to observe their cell growth effects by MTT colorimetry. Results The EGF exhibited a dose-dependent growth stimulating effect on U2 51MG cells, approaching the maximal effect at 10 ng ml, similar to the growth-promoting effect of 10% calf serum (P > 0.05); NC inhibited U2 51MG cell growth, and completely Block the effects of EGF. Conclusion Nicardipine can inhibit the growth of glioma cells and completely block the growth stimulatory effect of EGF, providing a new clue for clinical treatment of gliomas.
