目的研究逍遥散对氢溴酸东莨菪碱致阿尔茨海默病模型小鼠记忆功能及血清SOD、MDA、ChAT、AchE活性的影响,探讨逍遥散防治阿尔茨海默病的作用及其机制,阐释疏肝、养血、健脾三者结合防治阿尔茨海默病的配伍意义。方法60只SPF级KM小鼠随机分为空白组、模型组、逍遥散全方组、疏肝养血组、疏肝健脾组、养血健脾组等6组,每组10只。腹腔注射氢溴酸东莨菪碱复制阿尔茨海默病小鼠模型并分别以逍遥散全方及各配伍药组进行干预防治,给药第6、8天进行学习能力训练,第10天进行记忆功能测试,然后股动脉取血测定血清SOD、MDA、ChAT、AchE活性。结果逍遥散全方组、疏肝养血组、疏肝健脾组、养血健脾组皆可提高小鼠记忆潜伏时间,降低错误次数,提高血清SOD、ChAT活性,降低MDA、AchE活性,但记忆功能及血清SOD、MDA、ChAT、AchE的改善皆以逍遥散全方组最为明显。结论逍遥散能较好的改善阿尔茨海默病模型小鼠记忆功能、抗氧化能力及中枢神经递质活性,对阿尔茨海默病具有较好的防治作用,其疏肝、养血、健脾的结合配伍是有效防治阿尔茨海默病的组方结构。
Objective To investigate the effect of Xiaoyaosan on memory function, serum SOD, MDA, ChAT and AchE activity in Alzheimer’s disease model mice induced by scopolamine hydrobromide and to explore the role and mechanism of Xiaoyaosan in preventing and treating Alzheimer’s disease in mice. Liver, nourishing, spleen three combination of prevention and treatment of Alzheimer’s disease compatibility significance. Methods Sixty SPF KM mice were randomly divided into six groups (n = 10 in each group): blank group, model group, Xiaoyaosan Decoction group, Shugan Yangxue group, Shuganjianpi group, Yangxuejianpi group. Scopolamine hydrobromide was injected into mouse model of Alzheimer’s disease by intraperitoneal injection. Xiaoyaosan Decoction and various compatibilities were used to prevent and treat the mice. Learning ability training was performed on the 6th and 8th days, and memory test was performed on the 10th day , Then blood was collected from femoral artery to measure serum SOD, MDA, ChAT, AchE activity. Results Xiaoyao powder group, Shugan Yangxue group, Shuganjianpi group, Yangxuejianpi group can improve memory latency, reduce the number of errors, increase serum SOD, ChAT activity, reduce MDA, AchE activity, However, the improvement of memory function and serum SOD, MDA, ChAT and AchE were the most obvious in Xiaoyaosan Decoction group. Conclusion Xiaoyao powder can improve memory, antioxidant and CNS neurotransmitter activity in Alzheimer’s disease mice, and has a good preventive and therapeutic effect on Alzheimer’s disease in mice. Combination of spleen compatibility is effective to prevent and control the composition of Alzheimer’s disease.
