目的:研究柿叶黄酮(FLDK)对α-葡萄糖苷酶的抑制作用。方法:采用酶抑制剂筛选模型观察柿叶黄酮0.25~8.0 mg.mL-1对大鼠小肠α葡萄糖苷酶的作用,用人类结肠癌细胞株(Caco-2)细胞筛选模型观察柿叶黄酮0.1 mg.mL-1对麦芽糖酶、蔗糖酶的作用。结果:在酶抑制剂筛选模型体外实验中,FLDK对蔗糖酶、麦芽糖酶、乳糖酶活性的抑制作用呈浓度依赖性,当质量浓度达8.0 mg.L-1时,其对蔗糖酶、麦芽糖酶、乳糖酶的抑制作用均超过70%。在人类结肠Caco-2细胞中,FLDK对蔗糖酶、麦芽糖酶的抑制作用均超过75%。结论:FLDK对α-葡萄糖苷酶有较好的抑制作用,可用于防治糖尿病。
Objective: To study the inhibitory effect of FLDK on α-glucosidase. Methods: The enzyme inhibitor screening model was used to observe the effect of persimmon leaf flavonoids 0.25 ~ 8.0 mg.mL-1 on α-glucosidase in rat intestine. The human colon cancer cell line (Caco-2) Effect of mg.mL-1 on maltase and sucrase. Results: The inhibitory effect of FLDK on sucrase, maltase and lactase activities in a concentration-dependent manner in the in vitro experiments of enzyme inhibitors screening model, when the concentration of 8.0 mg.L-1, the effect of sucrosease, maltase , Lactase inhibition were more than 70%. In human colon Caco-2 cells, FLDK inhibited both sucrase and maltase by more than 75%. Conclusion: FLDK has a good inhibitory effect on α-glucosidase and can be used to prevent and treat diabetes.
