目的研究人牙周韧带细胞(periodontal ligament cells PDLCs)在溶胶-凝胶生物活性玻璃(sol-gel bioglass,SGBG)/聚羟基烷酸酯(poly-3-hydroxy butyrate-co-3-hydroxy valerate,PHBV)三维支架上的分泌功能,为牙周组织工程支架的选择提供依据。方法通过体外细胞培养实验,人牙周韧带细胞与SGBG/PHBV体外三维培养的直接接触法,测定上清液羟脯氨酸含量,检测材料对细胞分泌功能的影响。结果实验组上清液羟脯氨酸含量测定值与对照组有显著性差异,材料不影响细胞的分泌功能,同时支架材料的三维结构更能促进细胞的分泌功能。结论 SGBG/PHBV作为支架材料,有良好的生物相容性,有望应用于牙周组织缺损的组织工程修复。
Objective To study the effect of periodontal ligament cells (PDLCs) on the surface of solitary gel bioglass (SGBG) / poly-3-hydroxy butyrate-co-3-hydroxy valerate PHBV) three-dimensional scaffold secretion function, provide a basis for the selection of periodontal tissue engineering scaffold. Methods The content of hydroxyproline in supernatant was determined by in vitro cell culture experiment, direct contact method of human periodontal ligament cells with SGBG / PHBV in vitro culture, and the effect of materials on cell secretory function was tested. Results The content of hydroxyproline in the supernatant of the experimental group was significantly different from that of the control group. The material did not affect the secretion function of the cells, and the three-dimensional structure of the scaffold material further promoted the secretion function of the cells. Conclusion SGBG / PHBV as a scaffold material has good biocompatibility and is expected to be applied to tissue engineering repair of periodontal tissue defects.
