霍乱弧菌几种肠毒素基因的检测研究

目的 了解我省分离的霍乱弧菌中几种主要肠毒素基因的携带情况。方法 以地高辛标记基因探针 ,对我省历年分离的 91株霍乱弧菌 (包括EVC和VCO13 9)作霍乱肠毒素基因探针杂交检测。结果 EVC流行株和VCO13 9菌株中 ,普遍存在ctxAB、Zot、Ace等毒素基因 ,该 3种基因的携带率分别达到10 0 %,98 86 %,96 5 9%;而EVC非流行菌株则均不携带上述3种毒素基因。结论 提示EVC流行株和VCO13 9菌株具备较强的产毒能力 ,流行性强 ,应重点加强监测与研究。从ctxAB基因的RFLP杂交图谱分析 ,EVC流行株和VCO13 9在遗传特征上具相近的特点 ,结果均显示 2条阳性杂交带。 Objective To understand the carriage of several major enterotoxin genes in Vibrio cholera isolated in our province. Methods Digoxigenin-labeled gene probes were used to detect 91 strains of Vibrio cholerae (including EVC and VCO139) in our province as cholera enterotoxin gene probes. Results The virulence genes of ctxAB, Zot and Ace were common in EVC and VCO13 9 strains, and the carrier rates of these three genes were 100%, 98 86% and 96 59% respectively, while those of EVC non-endemic strains Do not carry the three toxin genes. The results suggest that EVC epidemic strains and VCO139 strains have strong ability to produce toxins, the epidemic, should focus on strengthening monitoring and research. From the RFLP map of ctxAB gene, EVC epidemic strains and VCO139 have similar genetic characteristics, and the results show that there are two positive hybridization bands.
