目的:为了探讨脑缺血再灌注外周血细胞因子介导的粒细胞趋化因子(CINC)水平的变化,采用动物实验方法对实验性脑缺血再灌注不同时间内外周血CINC和髓质过氧化物酶(MPO)活性的变化进行了观察。方法:选用改良线栓法复制Sprague-Dawley(SD)大鼠脑缺血再灌注实验模型。对不同时间内的血清MPO活性及CINC水平进行测定。结果:单纯缺血组MPO活性增高始于缺血6h并于缺血72h时达高峰,为0.15±0.01U/ml;CINC水平增高始于缺血1h,缺血6h时达高峰,为0.05±0.05ng/ml,以后逐渐下降并于缺血48h后恢复正常。缺血1小时再灌注组MPO活性增高始于再灌.注1h并于再灌注24h时达高峰,为0.42±0.12U/ml;CINC水平增高始于再灌注1h,再灌注3h时达高峰,为0.65±0.03ng/ml,以后逐渐下降并于再灌注24h后恢复正常。结论:实验性脑缺血再灌注外周血CINC有表达增高改变,其作用过程可能与白细胞激活过程有关,提示CINC增高可能在脑缺血再灌注损伤中起一定作用。
Objective: To investigate the changes of cytokine-mediated granulocyte chemotactic factor (CINC) in peripheral blood of cerebral ischemia-reperfusion rats. Animal experiments were conducted to evaluate the changes of CINC and medulla peroxidation in experimental cerebral ischemia-reperfusion Changes in enzyme activity (MPO) were observed. Methods: The experimental model of Sprague-Dawley (SD) rat cerebral ischemia-reperfusion was reproduced by modified thread method. Serum MPO activity and CINC levels were measured at different times. Results: The increase of MPO activity in ischemia group began at 6h after ischemia and peaked at 72h after ischemia, which was 0.15 ± 0.01U / ml. CINC increased from 1h after ischemia to 6h after ischemia, reaching 0.05 ± 0.05ng / ml, then gradually decreased and returned to normal 48h after ischemia. The increase of MPO activity in reperfusion group started at 1 hour after reperfusion and reached the peak at 0.24 ± 0.12U / ml at 1 hour after reperfusion. The level of CINC increased from 1 hour after reperfusion to 3 hours after reperfusion, 0.65 ± 0.03ng / ml, then gradually decreased and returned to normal after 24h reperfusion. Conclusion: The expression of CINC in peripheral blood of experimental cerebral ischemia-reperfusion rats may be increased. The possible mechanism may be related to the process of leukocyte activation, suggesting that increased CINC may play a role in cerebral ischemia-reperfusion injury.
