目的:观察黄芪有效组分对脂多糖诱导的大鼠肾小球系膜细胞表达血小板源性生长因子-BB(PDGFBB)的影响。方法:将黄芪有效组分(黄芪多糖、黄芪甲苷、毛蕊异黄酮)作用于经脂多糖刺激的大鼠肾小球系膜细胞(GMCs)后,采用逆转录-聚合酶链反应法、免疫细胞化学法检测PDGF-BB m RNA、蛋白的表达。结果:模型组经脂多糖刺激后PDGF-BB m RNA及蛋白表达增加(P<0.05),黄芪不同组分均能不同程度抑制PDGFBB m RNA及蛋白表达(P<0.05)。结论:说明黄芪多糖、黄芪甲苷、毛蕊异黄酮可不同程度抑制GMCs PDGF-BB m RNA及蛋白的表达,从而改善或延缓系膜增生性肾病肾小球的硬化。
OBJECTIVE: To observe the effects of active components of Astragalus on the expression of platelet-derived growth factor-BB (PDGFBB) induced by lipopolysaccharide in rat mesangial cells. Methods: Astragalus polysaccharide, Astragaloside IV and calycosin were treated with LPS in rat glomerular mesangial cells (GMCs), and then the expression of GST was detected by reverse transcription - polymerase chain reaction The expression of PDGF-BB m RNA and protein was detected by chemical method. Results: The mRNA and protein expressions of PDGF-BB and m RNA in the model group were significantly increased (P <0.05). Different fractions of Astragalus could inhibit the mRNA and protein expression of PDGFBB to varying degrees (P <0.05). Conclusion: Astragalus polysaccharide, Astragaloside IV and calycosin could inhibit the expression of PDGF-BB m RNA and protein in GMCs to varying degrees, so as to improve or delay the glomerular sclerosis of mesangial proliferative nephropathy.
