用油菜品种“西南302”(B.napes)为材料,研究了油菜种子贮藏蛋白的分离及部分性质。用 Na2HPO4-NaOH(pH 11.4)缓冲液抽提蛋白。 4%~20%连续梯度SDS—PAGE垂直板电泳表明,该贮藏蛋白组分十分复杂,其分子量范围是 8, 000~ 198,000,带型不仅多,而且含量很不一致,其中低分子量蛋白质(8, 000~ 30, 000)含量占 50%~ 60%以上。凝胶等电聚焦电泳表明,等电点范围为 pH3. 5~ 9,其中酸性蛋白质含量占 65%左右。
Using rapeseed variety “B.napes” as materials, the isolation and partial properties of rapeseed seed storage proteins were studied. The protein was extracted with Na2HPO4-NaOH (pH 11.4) buffer. The SDS-PAGE and vertical plate electrophoresis with 4% ~ 20% continuous gradient showed that the storage protein components were very complex with the molecular weight ranged from 8000 to 198000. There were not only more bands but also different contents. The low molecular weight proteins 8,000 ~ 30,000) content of 50% to 60% or more. Gel isoelectric focusing electrophoresis showed that the isoelectric point range was pH3. 5 ~ 9, of which acidic protein content accounted for about 65%.
