目的:探讨慢性再生障碍性贫血患者(cAA)血浆及CD3+、CD8+细胞胞质内干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)的表达情况及其在cAA发病机制中的作用。方法:应用ELISA法检测50例cAA患者及50例健康志愿者血浆IFN-γ、TNF-α的水平,流式细胞仪(FCM)检测外周血CD3+、CD8+细胞胞质内IFN-γ、TNF-α的表达水平。结果:cAA患者血浆中IFN-γ、TNF-α的水平显著升高,与正常对照组相比,差异有统计学意义(P<0.05),CD3+、CD8+细胞胞质内IFN-γ、TNF-α的水平亦显著升高,与正常对照组相比,差异有统计学意义(P<0.05)。结论:cAA患者T淋巴细胞高表达IFN-γ、TNF-α,其中CD8+T淋巴细胞亦高表达IFN-γ、TNF-α,这可能是cAA骨髓造血功能衰竭的机制之一。
Objective: To investigate the expression of IFN-γ and TNF-α in plasma and CD3 +, CD8 + cells of patients with chronic aplastic anemia (cAA) and its relationship with the pathogenesis of cAA In the role. Methods: The levels of plasma IFN-γ and TNF-α in 50 cAA patients and 50 healthy volunteers were detected by ELISA. The levels of IFN-γ and TNF-α in peripheral blood CD3 + and CD8 + cells were detected by flow cytometry (FCM) α expression level. Results: Compared with normal control group, the levels of IFN-γ and TNF-α in plasma of cAA patients were significantly increased (P <0.05). The levels of IFN-γ and TNF- α levels were also significantly increased compared with the normal control group, the difference was statistically significant (P <0.05). CONCLUSION: T lymphocytes of cAA patients overexpress IFN-γ and TNF-α. Among them, CD8 + T lymphocytes also overexpress IFN-γ and TNF-α, which may be one of the mechanisms of cAA bone marrow hematopoietic failure.
