目的了解急性早幼粒细胞白血病(APL)患者TCRVβ亚家族T细胞的分布及其克隆性。方法利用RT-PCR和基因扫描技术分析21例APL患者外周血单个核细胞的TCRVβ24个亚家族基因的CDR3长度,10例正常人作为对照。结果21例患者外周血T细胞所表达的Vβ亚家族数量差别很大,2~21个Vβ亚家族不等,其中,以Vβ(271.4%),Vβ1(561.9%)和Vβ(557.1%)出现频率较高,而Vβ10和Vβ14出现频率最低(14.3%)。除2例外,患者外周血均可检测到某些Vβ亚家族的克隆性T细胞,主要为Vβ12,Vβ21和Vβ23克隆性T细胞。结论APL患者外周血Vβ亚家族T细胞的倾斜性分布特点,并存在克隆性增生的T细胞,这可能是机体T细胞受白血病细胞相关抗原的刺激作用而引起机体产生相应的特异性免疫反应。
Objective To understand the distribution and clonality of TCR Vβ subfamily T cells in patients with acute promyelocytic leukemia (APL). Methods RT-PCR and gene scanning techniques were used to analyze the CDR3 length of TCR Vβ24 subfamily genes in peripheral blood mononuclear cells from 21 APL patients. Ten normal controls were used as controls. Results The number of Vβ subfamilies expressed in T cells in peripheral blood of 21 patients varied greatly, ranging from 2 to 21 Vβ subfamily, among which Vβ (271.4%), Vβ1 (561.9%) and Vβ (557.1%) appeared. The frequency was higher, and the frequency of Vβ10 and Vβ14 was the lowest (14.3%). With 2 exceptions, certain Vβ subfamilies of clonal T cells were detected in peripheral blood, mainly Vβ12, Vβ21, and Vβ23 clonal T cells. Conclusion The tilt distribution of Vβ subfamily T cells in peripheral blood of patients with APL, and the presence of clonally proliferating T cells, may be caused by the body’s T cells stimulated by leukemia cell-associated antigens.
