苦参碱(matrine)是提纯于中药苦参、山豆根等豆科槐属植物的一种生物碱活性成分。经现代研究证实其有多种药理作用,尤以其诱导人类白血病细胞分化引人关注。为探明其作用机制,根据其结构特点、理化性质,并结合研究需要,我们尝试用同位素催化置换法对苦参碱进行氚(3H)标记。标记结果:放射性总活度(RTA)>1.85×10Bq:放射性比活度(S)=10.06×10Bq/μg;放射性化学纯度(RCP)=95.2%。并对标记产物的理化性能及生物学效应作以鉴定。证明用氚标记苦参碱的方法是成功的,能满足进一步研究的要求。
Matrine is an alkaloid active ingredient purified from traditional Chinese medicine Sophora flavescens, Pied It has been confirmed by modern research that it has a variety of pharmacological effects, especially its induction of human leukemia cell differentiation. In order to explore its mechanism of action, according to its structural characteristics, physicochemical properties, and combined with research needs, we tried to carry out tritium (3H) labeling on matrine by isotope catalytic displacement. The result of the labeling is: total radioactivity (RTA)> 1.85 × 10Bq: specific radioactivity (S) = 10.06 × 10Bq/μg; radiochemical purity (RCP) = 95.2%. The physicochemical properties and biological effects of the labeled products were identified. It was proved that the method of labeling matrine with hydrazine was successful and could meet the requirements of further research.
