目的:探讨枳子提取物对实验大鼠肝组织基质金属蛋白酶-13(MMP-13)与金属蛋白酶组织抑制因子-1(TIMP-1)表达的影响。方法:SD大鼠48只,随机分为2组:正常对照组(16只)与模型组(32只),用四氯化碳制备大鼠肝纤维化模型,共造模6周,6周末各组分别处死大鼠8只,取肝脏进行HE染色;模型组大鼠随机分为3组:自然恢复组、对照组和给药组,于12周末处死各组大鼠,取肝脏,运用半定量逆转录聚合酶链反应(RT-PCR)检测肝组织中MMP-13 mRNA与TIMP-1 mRNA的表达。结果:MMP-13 mRNA的表达在各组之间并无显著性差异,而TIMP-1mRNA在各组之间有显著性差异,与正常组相比,自然恢复组大鼠肝组织TIMP-1 mRNA显著升高(P<0.05),而与自然恢复组相比,对照组和给药组大鼠肝组织TIMP-1 mRNA的表达显著降低(P<0.05),其中给药组TIMP-1 mRNA比对照组显著降低(P<0.05)。结论:枳子提取物逆转肝纤维化的机制可能是减少肝纤维化大鼠肝脏中TIMP-1mRNA的表达,逐渐恢复肝脏胶原降解系统,从而逆转肝纤维化。
Objective: To investigate the effects of Gardenia extract on the expression of matrix metalloproteinase-13 (MMP-13) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in experimental rats. METHODS: Forty-eight Sprague-Dawley rats were randomly divided into 2 groups: normal control group (16) and model group (32). Rat hepatic fibrosis model was prepared with carbon tetrachloride and a total of 6 weeks and 6 weeks were established. Eight rats were sacrificed in each group and the livers were used for HE staining. Model rats were randomly divided into three groups: natural recovery group, control group, and administration group. Rats in each group were sacrificed at the end of the 12th week and the livers were taken. Quantitative reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of MMP-13 mRNA and TIMP-1 mRNA in liver tissue. RESULTS: There was no significant difference in the expression of MMP-13 mRNA among the groups, but TIMP-1 mRNA was significantly different among the groups. Compared with the normal group, the TIMP-1 mRNA was observed in the liver tissue of the natural recovery group. Significantly increased (P<0.05), compared with the natural recovery group, the expression of TIMP-1 mRNA in the liver tissue of the control group and the administration group was significantly reduced (P<0.05), and the TIMP-1 mRNA ratio in the administration group The control group was significantly reduced (P<0.05). CONCLUSION: The mechanism of reversal of hepatic fibrosis by extracts of Gardenia jasminoides may be to reduce the expression of TIMP-1 mRNA in the liver of rats with hepatic fibrosis and gradually restore the collagen degradation system in the liver, thereby reversing hepatic fibrosis.
