目的建立高效毛细管电泳法(HPCE)同时测定杜仲中桃叶珊瑚苷、京尼平苷、松脂醇二葡萄糖苷、哈巴苷和绿原酸含量的方法。方法以60 mmol.L-1硼砂-20 mmol.L-1磷酸二氢钠-10%甲醇(pH 10.0)为电泳介质,未涂渍标准熔融石英毛细管(75μm×64.5 cm,有效长度56 cm)为分离通道,分离电压为20 kV,检测波长为210 nm,毛细管温度为25℃,压力进样为5 kPa×6 s。结果 5种指标成分的浓度与峰面积的线性关系良好(r>0.997 3);加样回收率为96.63%~103.73%,结果满意。结论该方法简单、准确,重复性较好,可用于杜仲药材或饮片的质量评价和控制。
OBJECTIVE To establish a method for the simultaneous determination of aucubin, geniposid, pinoresinol diglucoside, hababine and chlorogenic acid in Eucommia ulmoides by high performance capillary electrophoresis (HPCE). METHODS The standard fused silica capillary (75 μm × 64.5 cm, effective length 56 cm) was coated with 60 mmol·L-1 borax and 20 mmol·L-1 sodium dihydrogen phosphate-10% methanol (pH 10.0) To separate the channels, the separation voltage was 20 kV, the detection wavelength was 210 nm, the capillary temperature was 25 ° C and the pressure was injected at 5 kPa × 6 s. Results The linear relationship between the concentration of five index components and the peak area was good (r> 0.997 3). The recoveries of samples were 96.63% ~ 103.73%. The results were satisfactory. Conclusion The method is simple, accurate and reproducible. It can be used for the quality evaluation and control of Eucommiae medicinal herbs or Pieces.
