建立巴豆(Croton tiglium Linn)种仁中佛波醇的HPLC含量测定方法。采用岛津LC-2010型液相色谱仪,Diamonsil C18分析柱(250mm×4.6mm,5μm),流动相:乙腈-水(梯度洗脱),检测波长:234nm,流速:1mL/min,柱温:25℃。在选定色谱条件下佛波醇浓度在6.096—101.600μg/mL范围内与峰面积的线性关系良好,r=0.9999,平均加样回收率为96.40%,RSD为1.03%。本实验建立了RP-HPLC测定巴豆种仁中佛波醇的含量,为佛波醇的进一步研究提供了一种手段。
To establish a HPLC method for determination of phorbol in seeds of croton tiglium Linn. A Shimadzu LC-2010 liquid chromatograph was used with a Diamonsil C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase was acetonitrile-water (gradient elution). The detection wavelength was 234 nm and the flow rate was 1 mL / : 25 ° C. Under the selected chromatographic conditions, the phorbol has a good linear relationship with the peak area in the range of 6.096-101.600μg / mL with r = 0.9999, the average recovery was 96.40% and the RSD was 1.03%. In this experiment, the content of phorbol in croton kernels was established by RP-HPLC, which provided a means for the further study of phorbol.
