本研究利用高效液相色谱分析法测定经两种不同作用机制的拟精神病药(哌甲酯、氯安酮)长期作用后(6周)的大鼠海马游离氨基酸和皮层下组织多巴胺及代谢物的含量。结果发现,哌甲酯和氯安酮长期作用后大鼠海马的γ-氨基丁酸、甘氨酸、天门冬氨酸、游离氨含量降低;氯安酮还使谷氨酸增高、精氨酸降低。两种药物均有使皮层下组织多巴胺活动增高的趋势,且哌甲酯尤明显。
In this study, the free amino acids in the hippocampus and subcortical dopamine and its metabolites in rat hippocampus after long-term action (methylphenidate, chlordane ketone) by two different mechanisms of action were determined by high performance liquid chromatography Content. The results showed that the long-term effects of methylphenidate and chlordane ketone in rat hippocampus after γ-aminobutyric acid, glycine, aspartate, free ammonia content decreased; also increased glutamic acid ketone arginine, arginine decreased. Both drugs have increased subcortical dopamine activity trend, and particularly methylphenidate.
