从太平洋西加罗林群岛的帕劳(Palan)岛、莱恩群岛的范林(Fanning)岛、马歇尔群岛和夏威夷群岛采集了107种海藻,经冷冻或风干保存,用二种方法进行提取, 方法Ⅰ:在Waring匀浆器中,加入比样品重量5~10倍的有机溶剂,匀浆5分钟,混合物放置24小时或搅拌4~5小时,离心,上清液经真空蒸发,得粗脂溶性提取物。沉淀的残渣经干燥后用5~10倍量的水浸泡并搅拌4~5小时,放置24小时或更长的时间,离心,上清液冰冻干燥,得到水溶性提取物。水溶性提取物进行凝胶过滤(葡聚糖凝胶G-25),用0.1M碳酸氢铵洗脱,然后用阳离子交换层析(CM-葡聚糖凝胶C-25),活性成份以0.03~3.0M的醋酸铵梯度洗脱,于280nm处用紫外监测柱层析,得到初步纯化的流份。

The water-soluble extract was subjected to gel filtration (Sephadex G-25), eluted with 0.1 M ammonium bicarbonate, and then purified by cation exchange chromatography (CM-Sephadex C-25) 0.03 ~ 3.0M ammonium acetate gradient elution at 280nm with UV monitoring column chromatography to obtain a preliminary purification of the flow.
