以纤维素为载体,通过四氯化钛活化,可赋予纤维素较好的固酶功能。本文研究了固定化酶的制备条件和性质。当溶液5’-磷酸二酯酶的活力为200—300单位/毫升,经最佳条件固定化后,固定化5’-磷酸二酯酶的活力可达每克3000—3500单位(干基),酶的话力回收可达70—75%,固定化5’-磷酸二酯酶的热稳定性和贮藏稳定性与溶液酶相比有所提高。采用本法固定5’-磷酸二酯溶液酶具有载体制备简单易行、成本低廉、效果好、所用化工原料无毒等特点。
Cellulose as a carrier, through the activation of titanium tetrachloride, can give better enzyme immobilization of cellulose. This article studies the preparation conditions and properties of immobilized enzymes. When the activity of the solution 5’-phosphodiesterase is 200-300 units / ml, immobilized 5’-phosphodiesterase has the activity of 3000-3500 units per gram (dry basis) after being immobilized under optimal conditions, , The recovery rate of the enzyme is up to 70-75%, and the thermal stability and storage stability of the immobilized 5’-phosphodiesterase are improved compared with that of the solution enzyme. The method of immobilizing 5’-phosphodiester solution has the advantages of easy preparation, low cost, good effect and non-toxic chemical raw materials.
