[目的]检测朝鲜族人群Forkhead-box j1(Fox j1)基因的3个单核苷酸多态性(SNP)位点g.-460C>T,g.1805G>T,g.3375G>C的多态性,观察其与朝鲜族人群支气管哮喘易感性的关系.[方法]采用聚合酶链反应法和直接测定序列法检测32例朝鲜族哮喘患者和50例正常对照者的Fox j1基因的单核苷酸多态位点g.-460C>T,g.1805G>T,g.3375G>C的基因型,计算基因型频率和等位基因频率,进行Hardy-Weinberg平衡吻合度检验,并对比分析两组等位基因频率和基因型频率分布.[结果]Fox j1基因所选择的3个SNP位点在朝鲜族人群中均具有多态性,其基因型频率和等位基因频率分布在哮喘组和对照组中均符合Hardy-Weinberg平衡.哮喘组上述位点的基因型频率和等位基因频率与对照组比较差异均无统计学意义(P>0.05).[结论]未发现Fox j1基因的g.-460C>T,g.1805G>T,g.3375G>C的多态性与朝鲜族人群支气管哮喘易感性有显著相关性.
[Objective] To detect three single nucleotide polymorphisms (SNPs) of Fox J1 gene in Korean population g.-460C> T, g.1805G> T, g.3375G> C To investigate the relationship between Fox j1 gene polymorphism and susceptibility to bronchial asthma in Korean ethnic population. [Methods] Polymerase chain reaction and direct sequencing method were used to detect the single nucleotide polymorphisms of Fox j1 gene in 32 cases of Korean asthma patients and 50 normal controls The genotypes of nucleotide polymorphisms (g.-460C> T, g.1805G> T, g.3375G> C) were genotyped. The genotype frequency and allele frequency were calculated, and the Hardy-Weinberg equilibrium agreement was tested. The allele frequency and genotype frequency distribution of the two groups were analyzed. [Results] The three SNPs selected by Fox j1 gene were all polymorphic in Korean population. The genotype frequency and allele frequency were distributed in asthma Hardy-Weinberg equilibrium was found in the control group and the control group (P> 0.05). [Conclusion] The Fox j1 gene was not found in the asthma group The polymorphisms of g.-460C> T, g.1805G> T, g.3375G> C were significantly correlated with the susceptibility to bronchial asthma in Korean population.
