本实验采用化学合成法制备了五种定位氘标记的苯基羟乙酸酯类化合物,包括两个光学异构体。氘代丰度均在98%(原子百分)以上。 将氘标记的和非标记的苯基羟乙酸酯等摩尔混合后,配成1 mg/ml水溶液,分别对三组大白鼠ip给药,剂量为0.5 mg/(kg·d),连续2 wk,一组作空白对照。收集尿液,用Sep-Pak C_(18)小柱提取尿中CPG及其氘代化合物的代谢产物,应用气相色谱-质谱测定代谢产物。在气相色谱条件下难挥发的代谢产物,以薄层色谱分离
In this experiment, five kinds of deuterium-labeled Phenylglycolacetates were prepared by chemical synthesis, including two optical isomers. Deuterium abundances are 98% (atomic percent) or more. Deuterium-labeled and unlabeled phenyl glycolate were equimolar mixed to form a 1 mg / ml aqueous solution, respectively, three groups of rats ip administration, a dose of 0.5 mg / (kg · d), continuous 2 wk, a set of blank control. Urine was collected and the metabolites of CPG and its deuterated compound in urine were extracted with Sep-Pak C_ (18) cartridge. Metabolites were determined by gas chromatography-mass spectrometry. Metabolites that are less volatile under GC conditions are separated by thin-layer chromatography
