目的观察丹酚酸B(SalB)对小鼠脑缺血能量代谢的影响。方法NIH小鼠分为假手术组、缺血组、SalB组和尼莫地平(N im)组,测定脑组织线粒体膜电位(MMP)、脑能荷(EC)、磷酸肌酸(PCr)含量和Na+-K+-ATPase、Ca2+-ATPase的活性。结果脑缺血组脑缺血20、30 m in线粒体膜电位显著升高(P<0.01),SalB组和N im组可明显缓解脑缺血后线粒体膜电位的升高(P<0.01);脑缺血组脑组织EC、PCr的含量在缺血30 m in显著降低(P<0.05~0.01),SalB组和N im组可升高EC及PCr的含量(P<0.01);脑缺血组Na+-K+-ATPase和Ca2+-ATPase活性在脑缺血20、30m in时显著下降(P<0.01),而SalB组和N im组可缓解Na+-K+-ATPase、Ca2+-ATPase活性的下降(P<0.05~0.01)。结论SalB改善脑缺血能量代谢可能与其调整不同的环节有关。
Objective To observe the effect of salvianolic acid B (SalB) on energy metabolism of cerebral ischemia in mice. Methods NIH mice were divided into sham operation group, ischemia group, SalB group and nimodipine (N im) group. The contents of mitochondrial membrane potential (MMP), brain energy charge (EC) and creatine phosphate (PCr) in brain tissue were measured. And Na+-K+-ATPase, Ca2+-ATPase activity. Results The mitochondrial membrane potential was significantly increased at 20 and 30 m in cerebral ischemic group (P<0.01), and the mitochondrial membrane potential was significantly relieved in SalB group and N im group (P<0.01). The content of EC and PCr in the cerebral ischemic group was significantly decreased at 30 min of ischemia (P<0.05-0.01). The levels of EC and PCr in the SalB and N im group increased (P<0.01); cerebral ischemia The activities of Na+-K+-ATPase and Ca2+-ATPase were significantly decreased at 20 and 30 minutes after cerebral ischemia (P<0.01), while the activities of Na+-K+-ATPase and Ca2+-ATPase were decreased in SalB and Nim groups. P<0.05~0.01). Conclusion SalB can improve the energy metabolism of cerebral ischemia and may be related to different aspects of adjustment.
