目的研究酵母锌与姜黄素的联合作用对酒精性肝损伤的干预效果,为酒精性肝病的预防和治疗提供科学依据。方法将实验小鼠随机分为正常对照、酒精对照、姜黄素+低、高剂量酵母锌4组,梯度酒精暴露6周造酒精肝损伤模型,同时分别给予低、高剂量(2.5、10 mg/kg)酵母锌联合姜黄素(75 mg/kg)灌胃干预。测定血清丙氨酸转氨酶(ALT)、天冬氨酸转移酶(AST)水平,制作肝组织病理切片并测定肝组织总抗氧化能力(T-AOC)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、谷胱甘肽硫转移酶(GST)、谷胱甘肽过氧化物酶(GSH-PX)含量。结果姜黄素+低、高剂量酵母锌组ALT活力、MDA含量分别为(34.11±26.04)、(41.74±32.75)U/L和(0.99±0.19)、(0.97±0.25)nmol/mg prot,明显低于酒精对照组(81.54±37.56)U/L和(1.32±0.22)nmol/mg prot(P<0.05);姜黄素+高剂量酵母锌组GSH、GST、GSH-PX值分别为(1.03±0.15)、(13.84±2.72)、(37.56±4.67)mg/g prot,高于酒精对照组(P<0.05)。结论酵母锌与姜黄素联合对慢性酒精肝损伤具有良好的保护作用,其防护效应可能与其减轻氧化损伤有关。
Objective To study the effect of combined action of yeast zinc and curcumin on alcoholic liver injury and provide a scientific basis for the prevention and treatment of alcoholic liver disease. Methods The experimental mice were randomly divided into normal control, alcohol control, curcumin + low and high doses of yeast zinc 4 groups, alcoholic liver injury model induced by gradient alcohol exposure for 6 weeks and low and high doses (2.5, 10 mg / kg Yeast zinc combined with curcumin (75 mg / kg) intragastrically. Serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measured and histopathological sections of liver tissue were made. T-AOC, MDA, (GSH), glutathione S-transferase (GST) and glutathione peroxidase (GSH-PX) were measured. Results The ALT activity and MDA content in curcumin + low and high dose yeast zinc groups were (34.11 ± 26.04), (41.74 ± 32.75) U / L and (0.99 ± 0.19) and (0.97 ± 0.25) nmol / mg prot (81.54 ± 37.56) U / L and (1.32 ± 0.22) nmol / mg prot (P <0.05). The values of GSH, GST and GSH-PX in curcumin + high dose yeast zinc group were (1.03 ± 0.15), (13.84 ± 2.72), (37.56 ± 4.67) mg / g prot, higher than that of the alcohol control group (P <0.05). Conclusion Yeast zinc combined with curcumin has a good protective effect on chronic alcoholic liver injury, and its protective effect may be related to its reduction of oxidative damage.
