目的通过研究胃炎Ⅰ号对CAG大鼠胃黏膜MMP1 mRNA表达及酶活性机制的影响,探讨胃炎Ⅰ号对CAG大鼠胃黏膜损伤的疗效作用机制。方法采用胃炎Ⅰ号煎剂高、中、低剂量浓度及维酶素治疗慢性萎缩性胃炎大鼠,实时荧光定量PCR技术检测MMP1 mRNA表达,明胶酶谱法检测MMP1酶活性。结果与空白对照组比较,模型对照组大鼠胃黏膜MMP1 mRNA表达降低(P<0.05),MMP1酶活性升高(P<0.01);治疗后与模型对照组比较,中剂量浓度胃炎Ⅰ号对改善CAG大鼠胃黏膜病变疗效最佳,可提高MMP1 mRNA表达(P<0.05),降低MMP1酶活性(P<0.01)。结论在慢性萎缩性胃炎阶段,MMP1 mRNA表达水平降低,MMP1酶活性上升;提高MMP1 mRNA表达,降低MMP1酶活性,可能是胃炎Ⅰ号治疗慢性萎缩性胃炎的疗效机制之一。
Objective To study the effect of Weiyan No.1 on the expression of MMP1 mRNA in the gastric mucosa of CAG rats and the mechanism of the enzyme activity, and to explore the therapeutic mechanism of Weiyan No.1 on gastric mucosal injury in CAG rats. Methods The chronic, atrophic gastritis rats were treated with high, medium and low doses of Decoction No.1 and Weiwei I. The expression of MMP1 mRNA was detected by real-time fluorescent quantitative PCR and the activity of MMP1 was determined by gelatin zymography. Results Compared with the blank control group, the expression of MMP1 mRNA in gastric mucosa of the model control group was decreased (P<0.05), and the activity of MMP1 enzyme was increased (P<0.01). After treatment, compared with the model control group, the middle dose concentration of gastritis I was The effect of improving gastric mucosal lesions in CAG rats was the best, which increased MMP1 mRNA expression (P<0.05) and decreased MMP1 enzyme activity (P<0.01). Conclusion In the stage of chronic atrophic gastritis, the expression level of MMP1 mRNA is decreased and the activity of MMP1 enzyme is increased. Increasing the expression of MMP1 mRNA and decreasing the activity of MMP1 may be one of the therapeutic mechanisms of gastritis I in the treatment of chronic atrophic gastritis.
